毛果芸香碱对映体的毛细管电泳手性分离
Chiral Separation of Pilocarpine by Capillary Electrophoresis
【摘要】 采用 β_环糊精及其衍生物作为手性选择剂对毛果芸香碱对映体进行了分离 ,研究了环糊精类型和浓度对分离的影响 ,同时考察了背景电解质 pH、操作电压和温度等因素对对映体分离的影响 ;结果表明 ,采用羟丙基_β_环糊精 (HP_β_CD)可以使毛果芸香碱对映体达到基线分离 ,优化条件下手性分离度可达2.79,为此类药物提供了一种简便、快速的毛细管电泳手性分离分析方法。
【Abstract】 A chiral separation of pilocarpine is performed by capillary zone electrophoresis(CZE)on a fused silica capillary withβ-cyclodextrin(β-CD)or its derivatives as chiral selectors.The effects of the type and concentration of cyclodextrins used on resolution are studied.We also investigated the effects of other parameters on resolution,such as pH,applied voltage and temperature,and optimized the separaˉtion conditions.It was observed that a separation of pilocarpine(Rs=2.79)can be achieved by using50mmol/L phosphate buffer at pH2.5containing20mmol/L hydroxypropyl-β-cyclodextrin(HP-β-CD).
