构建重组乳酸乳球菌生产谷胱甘肽
Production of glutathione by recombinant Lactococcus lactis
【摘要】 以大肠杆菌染色体DNA为模板 ,分别扩增得到编码γ 谷氨酰半胱氨酸合成酶和谷胱甘肽合成酶的基因gshA和gshB。将gshA和gshB基因克隆到质粒pNZ814 8中 ,电转化乳酸乳球菌NZ90 0 0 ,获得重组菌NZ90 0 0(pNZ32 0 3)。在添加 10mmol L谷氨酸、半胱氨酸和甘氨酸的M17培养基中培养该重组菌 ,当OD6 0 0 达到 0 4时用乳酸链球菌素诱导 4h ,胞内谷胱甘肽含量达到 35 8nmol mg蛋白 (胞内浓度相当于 14 0mmol L) ,这是在革兰氏阳性菌中生产谷胱甘肽的首例报道。
【Abstract】 The genes gshA and gshB encoding γ glutamylcycteine synthetase and glutathione synthetase were amplified from the chromosomal DNA of Escherichia coli TG1, respectively, and then inserted into plasmid pNZ8148 to get a hybrid plasmid The resulting plasmid pNZ3203 were introduced into Lactococcus lactis subsp cremoris NZ9000 by electroporation An incredibly high intracellular content ofGSH, 358 nmol/mg protein, was achieved after induction of nisin for 4 hours In L lactis NZ9000 (pNZ3203), the levels of GSH upon addition of 5 mmol/L of cysteine appeared to be approximately 6 fold that of the level without addition of cysteine
