运用生物信息技术分析日本血吸虫Z88基因cDNA序列
Sequence Analysis of Schistosoma japonicum Z88 cDNA by Bioinformatics Method
【摘要】 目的 通过基因序列分析 ,寻找日本血吸虫新的基因 ,为日本血吸虫病防治筛选新的候选疫苗抗原。方法 从已构建的日本血吸虫成虫 c DNA文库中挑取已知克隆 (克隆号为 Z88) ,测定其全长 c DNA序列后 ,再运用生物信息学方法以及相关软件对此基因进行分析。结果 通过测定以及分析可知此基因的 c DNA序列为1 3 0 3 bp,编码 3 49个氨基酸 ,为 Dna J-相似蛋白 ,与果蝇、鲐、人及鼠的 Dna J-相似蛋白有一定同源性。二级结构分析预测提示其具有一定抗原性。结论 Z88基因为日本血吸虫 Dna J-相似蛋白基因 ,可能参与蛋白折叠、装配和运输等功能
【Abstract】 Objective To discover new candidate vaccine antigen for schistosomiasis control by analysis of the squence structure of the cDNA of Schistosoma japonicum. Method The Z88 cDNA sequance acquired from the cDNA library of Schistosoma japonicum was analysed by bioinformatics method. Results The overall length of Z88 cDNA fragment was 1 303bp and coded 349 amino acid residues. It was homology to DnaJ-like protein of Drosophila melanogaster, Danio rerio, Homo sapiens and Mus musculus with identity of 38.44%, 35.60%, 35.95% and 35.04%. Secondary structure analysis showd that this protein had antigenicity. Conclusion The Z88 cDNA is the gene of Schistosoma japonicum DNAJ-like protein which would probably be the molecule participating in the function of polypeptide fold, assemble and transportation.
