应用分子技术验证中毒性休克综合征分离菌
Identification of Toxic Shock Syndrome Isolated Bacteria with Molecular techniques
【摘要】 目的 应用分子技术对江苏省 1990至 1995年中毒性休克综合征分离菌进行检测。方法 随机取 5株 (每年 1株 )分离菌进行 16S核糖体RNA基因 ( 16SrRNA基因或 16SrDNA)测序 ,与基因库中菌种比较同源性 ,从基因水平上确定细菌种类。分别将 5株分离菌的基因组经 2 0USamⅠ酶切消化后经脉冲场凝胶电泳(PFGE) ,确定其相关性。结果 16SrRNA基因测序分析证明分离菌均为缓症链球菌 ,5株细菌基因组经 2 0USamⅠ酶切消化后PFGE谱完全一致。结论 江苏省中毒性休克综合征流行疫情是由缓症链球菌引起的。
【Abstract】 Purpose To identify the bacteria in Jiangsu Province of China from 1990 to 1995 based on molecular techniques. Methods Five bacterial isolates were randomly analyzed(one each year)for homogeneities by comparison with 16S rRNA gene sequences in the genebank. The extracted genomic DNA was digested with the 20U Sam Ⅰ enzyme respectively,then their interrelationships were performed according to the pulsed field gel electrophoresis(PFGE). Results Five isolates from patients of 1990 to 1995 identified as S.mitis based on 16S rRNA gene sequencing and display indistinguishable PFGE patterns after digestion with 20U Sam Ⅰ. Conclusions These data demonstrated epidemiological relatedness of the bacterial isolates in Jiangsu Province of China,and suggested spread of S.mitis related streptococcus toxic shock syndrome(STSS)outbreak.
