上海地区汉坦病毒基因分型和序列分析

【摘要】 [目的 ] 研究上海地区汉坦病毒基因型分布。  [方法 ] 采用逆转录聚合酶链反应 (RT -PCR)、核苷酸测序法和系统发生树分析上海宿主动物中获得的 3 6株汉坦病毒M基因片段。  [结果 ] 上海郊区汉坦病毒流行株有HTN型和SEO型 ,以HTN型为主 ;市区流行株为SEO型。核苷酸序列同源性及系统发生树分析表明上海地区HTN型汉坦病毒存在 3个不同分支 ,不同HTN型病毒间的核苷酸的差异在 6% -19%。  [结论 ] 上海地区汉坦病毒以HTN型为主。可能存在汉坦病毒的不同基因亚型 ,对本市肾综合征出血热 (HFRS)的防治有重要指导意义

【Abstract】 Objective To find out the distribution of hantavirus genotypes in Shanghai. [Methods] The M segments of 36 strains of Hantavirus isolated from host animals with HFRS in Shanghai were detected by RT-PCR and sequenced. The obtained sequences were then analysed by phylogenetic tree software. [Results] The Hantavirus strains of HTN and SEO, mainly HTN, existed in the suburb of Shanghai, while the strain of SEO existed in the urban area of Shanghai. Homological and phylogenetic analysis suggested 3 different subtypes belonged to a novel subgenotype of HTN existed in Shanghai. The differences of partial M nucleotide sequence between the subtypes of HTN viruses were 6%-19%. [Conclusion] The strain of Hantavirus in Shanghai mainly belongs to HTN. The different subtypes of Hantavirus may exist in Shanghai, which is valuable for the prevention and control of HFRS.
