姜黄素对食高脂饲料家兔血管内皮功能的保护作用
Protective effect of curcumin on endothelial function in cholesterol fed rabbits
【摘要】 目的 观察姜黄素对高脂血症家兔血管内皮功能的保护作用。方法 36只雄性新西兰大白兔随机分为3组 ,即对照组 (喂普通饲料 )、模型组 (喂普通饲料 +1 5 %胆固醇、5 %的猪油 )和姜黄素组 (喂普通饲 +1 5 %胆固醇、5 %的猪油 +姜黄素 5 0mg·kg-1·d-1)。试验 4周后采血行血脂分析 ,酶法测NO ,放免法测ET 1,Elisa法测vWF ;采用离体血管功能实验方法 ,分别观察预先用苯肾上腺素作为收缩剂处理后的各组降主动脉血管环对乙酰胆碱和硝酸甘油的舒张反应。结果 姜黄素组血清TC、LDL C、TG及ET 1、vWF水平明显下降 ,NO水平明显增高 ,与模型组比较差别显著性(P <0 0 5 ) ;与对照组比较 ,模型组降主动脉血管环对乙酰胆碱 (3× 10 -7~ 3× 10 -5mol/L)刺激的舒张反应下降 ;姜黄素组血管环对乙酰胆碱刺激的舒张反应明显改善 ,与模型组比较有显著差别。三组血管环对不同浓度的硝酸甘油刺激的舒张反应无差异 (P >0 0 5 )。结论 姜黄素能纠高脂血症家兔内皮血管活性物质的紊乱 ,改善内皮依赖性舒张功能 ,对血管内皮功能有保护作用。
【Abstract】 Objectives To investigate the effects of curcumin on the dysfunction of endothelial cell during the progression of atherosclerosis in rabbits fed with choleslerol-rich diet. Methods 36 New Zealand white rabbits were randomed into 3 groups: control group was fed normal chow, model group was fed 1.5% cholesterol and 5% fat chow, curcumin group was given 1.5% cholesterol and 5% fat chow plus curcumin 50 mg·kg -1·d -1. Three groups were fed for 4 weeks. After experiment, serum lipid, nitric oxide (NO) content and plasma endothelin-1 (ET-1), von willebrand factor (vWF) concentrations were assayed. Descending aortae were harvested and cut into 2 mm rings. Rings then were immersed in cold krebs solution. These rings were progressively stretched for 2 hours. Until the optimal length-tension. Relation was achieved as assessed by contraction to phenylephrine (Phe). Then relaxation of aortae rings to acetylcholine (Ach) and nitroglycine were measured respectively. Results In curcumin group, serum TC,LDL-C,TG level, ET-1 concentration and vWF content decreased, NO concentration increased as compared with model group(P<0.05). Acetylcholine caused dose-dependent relaxation in aortae rings in three groups. In model group, relaxation of aortae rings to Ach(3×10 -7~3×10 -5) was significantly decreased compared with control group (P<0.05). The response of aortae rings to Ach were improved in curcumin group, there was a significant difference between curcumin and model group (P<0.05). Nitroglycine also caused dose-dependent relaxation in aortae rings, but there was no significant difference among control group,model group and curcumin group. Conclusions The curcumin can protect endothelial function of cholesterol-fed rabbits.
