人脑胶质瘤EGFRAKT通路活性的研究

【摘要】 目的 :研究胶质瘤中细胞EGFR和磷酸化AKT(p AKT)的表达以及EGFR与AKT活性的相互关系。方法 :用免疫组化方法观察 6例正常脑组织、5 0例胶质瘤标本和 2个恶性胶质瘤体外细胞系的EGFR表达 ,并应用Westernblot分析p AKT的表达。结果 :EGFR在胶质瘤中的表达阳性率随肿瘤恶性程度增加而增加 ;其蛋白表达水平与病理分级呈正相关。Westernblot分析发现在正常脑组织中p AKT蛋白水平表达极低 ,胶质瘤中p AKT的表达水平均随肿瘤恶性程度增加而明显上升 ,且与胶质瘤病理级别正相关 ;低恶度肿瘤与高恶度肿瘤有明显差异。EGFR的阳性表达率与AKT磷酸化水平显著相关。结论 :EGFR AKT通路的活性状态在胶质瘤的恶性进展具有重要作用 ,该通路可能是胶质瘤恶性表型的责任通路。

【Abstract】 Aim:To study the activation of EGFR-PI3K-AKT pathway in gliomas.Methods:EGFR expression was detected in six normal brain tissue samples, fifty glioma samples and two glioblastoma cell lines with immunohistochemical methods, phosphorylated-AKT(p-AKT) expression was studied by Western blot.Results:The positive rate of EGFR was increased with the ascending of tumor grade. Nearly visible expression of p-AKT could be found in six normal brain tissues by Western blot. However,the expression level of p-AKT increased significantly with the ascending of tumor grade, and the expression level in grade Ⅲ and Ⅳ was higher than that in low-grade gliomas. The expression rate of EGFR correlated well with p-AKT level.Conclusion:Activation of EGFR-PI3K-AKT pathway might contribute to the malignant progress of gliomas, this pathway could be an important one responsible for the development of malignant phenotype of gliomas.
