三种中药复方对慢性束缚应激大鼠行为及皮层和海马NT3的影响

【摘要】 目的 研究慢性束缚应激时大鼠行为学变化、皮层和海马CA1区神经营养蛋白 3(NT3)的变化以及逍遥散、四君子汤、金匮肾气丸对其的影响。方法 用特制束缚架连续束缚 7d与 2 1d、每天 3h的方法制作大鼠束缚应激模型 ,用免疫组织化学方法结合图像分析检测大鼠皮层和海马CA1区NT3的变化。结果  2 1d模型组大鼠中央格停留时间明显延长 ,穿格次数明显减少 ,7d模型组大鼠的穿格数减少 ;两个模型组大鼠的挣扎次数明显减少。 3个复方都能缩短中央格停留时间 ,增加穿格次数和挣扎次数。连续束缚 7d后大鼠大脑皮层的NT3开始下降 ,至 2 1d模型组与正常对照组相比均有显著下降 (P <0 0 5 )。连续束缚 7d、2 1d后大鼠海马CA1区的NT3与正常对照组相比积分光密度明显降低 (均P <0 0 5 )。 3个复方均能升高皮层和海马NT3的积分光密度和海马中NT3的阳性细胞数 ,逍遥散能升高皮层NT3的阳性细胞数。结论 皮层和海马CA1区NT3下降参与了慢性应激的变化 ,疏肝、健脾、补肾的中药复方均能改善大鼠的行为变化 ,能上调NT3;但以逍遥散的调节作用较强 ,优于四君子汤和金匮肾气丸

【Abstract】 Objective To observe the changes in the behavior and NT 3 in the cortex and hippocampal CA 1 region in the rats irritated by the chronic stressor of restraint, and to investigate the effects of 3 TCM formulas, namely Xiaoyao Powder, Sijunzi Decoction and Jinguishenqi Pills, on the rats. Methods The rat model of chronic stress to restraint was induced by restraining the rat on a specially-designed restraint rack 3 hours each day for 7 and 21 days, respectively. The behavior of the model rats was observed and the effects of the 3 TCM formulas on NT 3 in the cortex and hippocampal CA1 region were detected by the immunohistochemical method and image analysis. Results The duration of staying at the central grille was obviously longer and the times of passing through the grille were obviously decreased in the rats undergone 21 days of irritation by the chronic stressor of restraint; the times of passing through the grille were decreased in the rats undergone 7 days of the irritation; and the times of struggling in the rats in both model groups were obviously decreased. All the 3 TCM formulas could shorten the duration of staying at the central grille, and increase the times of passing through the grille and the times of struggling. The level of NT 3 in the cortex in the model rats began to decrease after 7 days of the irritation and the decrease became significant after 21 days of the irritation, as compared with that in the control rats ( P <0.05). The integral optical density of NT 3 in the hippocampal CA1 region in the rats in both model groups was significantly decreased, as compared with that in the control rats ( P <0.05). All the 3 TCM formulas could increase the integral optical density of NT 3 in both cortex and hippocampus and the number of hippocampal NT 3 positive cells; and Xiaoyao Powder could increase the number of cortical NT 3 positive cells. Conclusion The decrease in the level of NT 3 in the cortex and hippocampal CA 1 region is related with the reaction caused by the chronic stressor of restraint, and Xiaoyao Powder, Sijunzi Decoction and Jinguishenqi Pills all can improve the behavior of the model rats and upregulate the level of NT 3, among which Xiaoyao Powder shows a stronger effect.
