小柴胡汤治疗大鼠子宫内膜异位症的实验研究

【摘要】 目的:采用大鼠子宫内膜异位症(EMS)动物模型,探讨中药制剂小柴胡汤(XCHT)对EMS异位内膜的作用。方法:通过透射电镜技术观察XCHT对异位内膜细胞形态结构的影响。结果:中、高剂量XCHT10、15g/kg·d-1组对异位内膜有明显抑制作用,异位内膜体积明显减小(P<0 01),低剂量XCHT5g/kg·d-1组体积也有减小(P<0 05),而对照组体积无明显减小(P>0 05)。透射电镜下可见中、高剂量XCHT组治疗后较多腺上皮细胞出现凋亡的特征,表现为细胞体积变小、核固缩、胞浆和核染色质凝集、密度增高,细胞见凋亡小体,同时间质细胞中也可见一些坏死细胞。结论:XCHT对EMS大鼠异位内膜有明显抑制作用,其作用机制可能是通过调整宿主局部环境中的免疫状态和功能,而诱导细胞的凋亡和坏死。

【Abstract】 Aim: Effects of Xiao Chai Hu Tang(XCHT) on experimental endometiosis in female rats were evaluated. Methods: Measuring the ectopia endometrium volum and observing ultrastructural change by electron microscopy. Results: After the treatment with middle or high-concentraction (10、15 g/kg) XCHT, it could markedly inhibit endometrial explant growth (P<0.01). However, low-concentraction XCHT (5 g/kg) indistinctively inhibit it (P<0.05). The explant volume of control group did not markedly minish (P>0.05). Meanwhile, the glandular cell in the endometrial explant after therapy with middle or high-concentraction XCHT showed characteristic features of apoptosis which displayed the cell decreased in size, karyopyknosis, cytoplasmic and nuclear chromatin condensation, density increased and a lot of apoptotic bodies among cell. Whereas some stromal cell displayed degeneration and necrosis.Conclusion:The results suggested that XCHT had effects of inhibiting ectopia endometrium by inducing cell death, including apoptosis and necrosis.
