【摘要】 目的　比较多发性骨髓瘤 (MM)来源的树突状细胞 (DC)与正常人来源DC在形态、表型的差异 ,为主动特异性免疫治疗MM提供依据。方法　获取MM患者及正常人外周血单个核细胞 ,应用无血清培养基 (AIM V) ,加入GM CSF、IL 4、TNF α诱导DC产生 ,并分别比较两种来源DC在光、电镜及免疫表型上有无差异。结果　通过光、电镜观察DC的形态并用流式细胞仪检查细胞表面标志CD1a、CD83、CD80、CD86、HLA DR、CD14 ,统计学分析比较MM患者与正常人来源的DC表型 ,差别无显著意义 (P >0 .0 5 )。结论　应用无血清培养基可成功地培养出DC ,MM患者来源的DC在形态、表型上与正常人无明显差异更多还原

【Abstract】 Objective To compare the morphology and phenotype differences of dendritic cells DCs from mutiple myeloma(MM) patients and normal donors and provide the evidence of initiative and specific immunotherapy of MM.Methods Peripheral blood mononuclear cells were isolated from MM patients and normal donors and were cultured in serum-free medium and in the presence of GM-CSF,IL-4 and α-TNF to induce DCs’production.Compared the differences of the two kinds DCs in microscope,electron microscope and flow cytometry.Results Cells morphology and surface mark CD1a,CD83,CD80,CD86,HLA-DR and CD14 were analyzed from MM patients and normal donors.No obvious significance was observed(P>0.05).Conclusion DCs from MM patients has no obvious difference from normal donors.更多还原
