榆叶梅和海棠花粉萌发率的测定及超低温保存方法
Studies on pollen germination rate and cryopreservation method in Prunus triloba and Malus spectabilis
【摘要】 研究了榆叶梅和海棠花粉萌发所需的最佳蔗糖浓度和温度;分析了不同贮藏方法和不同解冻温度对花粉萌发率的影响。结果表明:榆叶梅和海棠的花粉适宜发芽温度为20℃,培养基的适宜蔗糖浓度为10%;超低温(-196℃)保存后的萌发率最高;超低温保存中海棠花粉以自来水冲洗解冻效果最好;榆叶梅花粉则以室温下解冻效果最好。
【Abstract】 The pollen of Prunus triloba and Malus spectabilis were studied on the optimal sucrose concentration,temperature of pollen germination,and infects on p ollen germination rate under different storage methods and different thawed temp eratures. The result indicated that the optimal pollen germination temperature i s 20℃ and the optimal sucrose concentration of medium is 10%. The rate of polle n germination is highest in cryopreservation. The pollen of Malus spectabilis th awed effect through tap water is the best in cryopreservation,and that of Prunus triloba pollen is best in room temperature.
