丹参酮ⅡA对瘢痕成纤维细胞增殖及凋亡的影响

【摘要】 目的 研究天然药物丹参酮ⅡA对瘢痕成纤维细胞的作用 ,以期寻找新的治疗增生性瘢痕的有效药物。方法 以四甲基偶氮唑 (MTT)法和特异性荧光染色及原位末端标记技术检测丹参酮ⅡA对成纤维细胞增殖及凋亡的影响。结果 丹参酮ⅡA对成纤维细胞的增殖具有显著的抑制作用 ,且呈浓度和时间依赖性。采用荧光染色和TUNEL法检测凋亡指数 ,均呈剂量依赖性增高 (P <0 0 1)。结论 丹参酮ⅡA对瘢痕成纤维细胞的增殖具有显著的抑制作用 ,并且能够诱导其发生凋亡 ,为寻找有效治疗增生性瘢痕的药物提供了新的思路。

【Abstract】 Objective To study the effect of herbal medicine Tan IIA on scar-derived fibroblasts and to explore new drug for human pathological scar. Methods Fibroblasts were cultured as an experimental model and the effect of Tan ⅡA were studied. Cell proliferation was determined by MTT assay, and apoptosis of fibroblasts induced by Tan ⅡA were analyzed by fluorescent staining and TUNEL. Results Tan ⅡA significantly inhibited the proliferation of fibroblasts, and significantly induced apoptosis of cultured fibroblasts by Hoechst 33258/PI staining and by counting TUNEL-positive cells(P<0.01). Conclusion Tan ⅡA can significantly inhibit the proliferation and induce the apoptosis of cultured fibroblasts. It may be developed as a new drug for the treatment of pathological scar.
