【摘要】 目的　应用彗星实验技术观察脱氧雪腐镰刀菌烯醇 (DON)对Vero细胞DNA损伤与修复特征 ,探讨DON的遗传毒性。方法　① 1μmol、5 μmol、10 μmolDON对指数生长期的Vero细胞分别染毒 4、16h后 ,观察DNA的损伤。② 10μmolDON处理的细胞重新孵育 15、3 0、60、12 0min ,观察DNA损伤修复。③以彗星实验技术结合“彗星图像分析软件(KIAS)”获得彗星细胞主要参数 ,并进行统计分析。结果　①随染毒剂量增加 ,染毒时间延长 ,受损伤细胞的数量逐渐增加 ,细胞DNA损伤程度逐渐加重 ;短时间染毒 ( 4h) ,损伤以DNA碎片的数量增加为主 ;较长时间染毒 ( 16h) ,损伤以DNA碎片变小为主。②去除DON ,重新孵育若干时间 ,随修复时间延长 ,受损伤细胞数量逐渐减少 ,细胞DNA损伤程度逐渐减轻 ,提示受损细胞在进行自我修复 ;短时间染毒 ( 4h)修复启动时间较短 ( <15min) ,而长时间染毒 ( 16h)修复启动时间也相应延长 ( >15min) ;孵育 12 0min后 ,修复完成。结论　DON毒素可致Vero细胞DNA损伤 ,重新孵育若干时间后损伤可修复。更多还原

【Abstract】 Objective To assess the genetic toxicity of deoxynivalenol(DON)with Comet assay through studying the DNA damage and repair of Vero cell induced by DON.Methods ①Vero cell in exponential growth period was treated with DON of 1 μmol、5 μmol、10 μmol,for 4 and 16 hours,respectively,to observe the damage of DNA.②The cells induced by 10 μmol DON were reincubated for 15,30,60 and 120 minutes,respectively,to investigate the repair of DNA.③The main parameter were obtained by combination of Comet assay and Komet Image Analysis System(KIAS),and the data was analyzed by statistic software SPSS.Results ①With the dose and time increasing,the damaged cells increased,and the damage of DNA was serious;In short-time administration(4 h),the main damage was the increase of DNA fragment;In long-time administration(16 h),the main damage was that DNA fragment became smaller.②Removing DON,with the repair time prolonging,damaged cells reincubated for some time decreased,and the damage of DNA lessened.This showed that damaged cells could repair themselves;cell repair in short-time administrated was earlier(<15 min)than that in long-time administration(16 h)(> min);After incubating for 120 minutes ,the repair completed.Conclusions DON toxin can damage the DNA of Vero cell,and the damage can be repaired after reincubating for some time.更多还原
