乌鸡白凤丸有效成分对Aβ1-40诱导的电压门控性钙通道电流增强作用的影响
INHIBITORY EFFECT OF BFP ON THE INCREASED VOLTAGE-GATED CALCIUM CHANNELS CURRENTS INDUCED BY Aβ1-40
【摘要】 ①目的 研究β 淀粉样蛋白 (Aβ1 40 )对大鼠肾上腺髓质嗜铬细胞瘤 (PC1 2 )细胞电压门控性钙通道(VGCC)电流的影响 ,以及乌鸡白凤丸有效成分 (BFP)对此效应的干预作用。②方法 将PC1 2细胞分成空白对照组、Aβ1 40 组、BFP组、Aβ1 40 +BFP组 ,分别应用培养液、含 1 μmol/LAβ1 40 的培养液、含 1 0 0mg/LBFP的培养液以及含 1 μmol/LAβ1 40 和 1 0 0mg/LBFP的培养液进行孵育 6h。在膜片钳全细胞记录模式下 ,记录各组细胞VGCC电流峰值 ,计算细胞的电流密度 (电流值 /膜电容 ) ,作为Ca2 + 内流指标。③结果 Aβ1 40 组VGCC平均电流密度为(4 5 .2 6± 2 9.2 9) pA/pF ,对照组为 (2 3.0 0± 2 1 .35 ) pA/pF ,两组相比 ,差异有极显著性 (t =2 86 ,P <0 .0 1 ) ;Aβ1 40 +BFP组的平均电流密度为 (1 2 .6 7± 1 1 .5 5 ) pA/pF ,与Aβ1 40 组比较 ,差异有显著意义 (F =2 9.32 6 ,q =7.5 5 4 ,P <0 .0 1 )。④结论 Aβ1 40 能够促进VGCC开放 ,使Ca2 + 内流增多 ,引起细胞损伤 ;BFP能有效抑制Aβ1 40 所引起的VGCC电流增强效应 ,在一定程度上避免了细胞内钙超载 ,从而阻止Aβ1 40 的毒性作用
【Abstract】 Objective To study the effects of amyloid β-protein(Aβ 1-40 ) on the voltage-gated calcium channels(VGCC) currents of PC12 cells and the protective action of Bak Foong Pills(BFP). Methods PC12 cells were divided into four groups: Control Group, Aβ 1-40 -treated Group, BFP-treated Group, Aβ 1-40 and BFP-treated Group, and the cells in each group were incubated in different cultures for six hours. Currents were measured by whole-cell patch-clamp recording and current density was obtained by current-amplitude/ membrane capacitance as the marker of the Ca 2+ influx. Results The VGCC average current density of the Aβ 1-40 -treated Group and Control Group was (45.26±29.29)pA/pF and (23.00±21.35)pA/pF, respectively. A significant difference existed between the two groups (t=286, P <0.01). The average current density of the Aβ 1-40 and BFP-treated Group was (12.67±11.55)pA/pF, which was significantly different from that of the Aβ 1-40 -treated Group( F=29.326,q=7.554,P <0.01). Conclusion Aβ 1-40 can increase the VGCC currents of PC12 cells, which results in the increased Ca 2+ influx and cell injuries. BFP can effectively inhibit this effect and prevent the toxicity of Aβ 1-40 by the avoidance of Ca 2+ overload.
