【摘要】 目的 :探讨右旋柠烯 (D limonene)对不同类型人胃癌细胞的生长抑制作用及其机制。方法 :以D limonene作用于SGC 790 1、BGC 82 3及AGS三种不同类型人胃癌细胞 ,采用噻唑蓝 (MTT)比色法、流式细胞仪及电镜检测细胞生长抑制率、凋亡率以及形态学改变 ,免疫细胞化学法检测 p5 3、bax和bcl 2基因表达。 结果 :D limonene处理后的细胞生长抑制率与凋亡率均增加 ,与药物浓度呈正相关。细胞呈现核固缩、染色质边集、凋亡小体形成等典型凋亡表现。经D limonene处理后SGC 790 1、BGC 82 3细胞株 p5 3、bax蛋白表达明显增加 ,bcl 2蛋白表达降低。AGS细胞株bax蛋白表达明显增加 ,bcl 2蛋白表达降低。 结论 :D limonene对人胃癌细胞的抑制作用主要是通过诱导细胞凋亡 ,不同类型的细胞株作用途径可能不同。升提 p5 3与bax及降低bcl 2的蛋白表达为其诱发胃癌细胞凋亡的机制之一。更多还原

【Abstract】 Objective: To investigate the growth-inhibition effects of D-limonene on different gastric cancer cell lines. Methods: Three human gastric cancer cell lines were selected as object of study. MTT, electron microscopy, flow cytometry were used to detect the growth-inhibition rate, apoptotic index and morphologic changes; immunocellchemistry method was used to detect the gene expression of p53, bax and bcl-2. Results: Compared with control group, the growth-inhibition rate, apoptotic index were significantly increased with increasing D-limonene content and typical characters of apoptosis were observed in D-limonene treated group. The gene expression of p53 and bax were increased apparently in SGC-7901,BGC-823 cell line treated with d-limonene but not in AGS cell line. While the expression of bcl-2 were decreased significantly in all three cell lines. Conclusion: D-limonene can inhibit the growth of human gastric cancer cell mainly through inducing cancer cell’s apoptosis by cell line specific way. Increasing the expression of p53 and (or) bax, decreasing the expression of bcl-2 may be one of the mechanisms.更多还原
