【摘要】 通过反转录—聚合酶链反应 (RT— PCR)扩增了猪轮状病毒国内地方分离株 JL 94的 VP6基因 c DNA片段。将其插入克隆载体质粒 p MD18- T的 Eco RV酶切位点处 ,构建了重组质粒 p MD18- T— VP6。对克隆的 VP6基因进行序列测定 ,测序结果显示 VP基因全长 135 6 bp,并与猪 A群轮状病毒 OSU(亚组 )、Gottfried(亚组 )的 VP6进行同源性比较 ,结果显示 ,核苷酸序列同源性分别为 86 .85 %、82 .4 1% ,氨基酸序列同源性分别为 97.4 8%、93.2 0 % ,结果表明 JL 94分离株与 OSU株在基因型上更为接近更多还原

【Abstract】 The porcine rotavirus JL94 isolate was propagated in MA104 cells and its dsRNA was extracted.Its cDNA segments of complete gene 6 were obtaied from RT-PCR and cloned into pMD18-Tvector and sequenced.The results of sequencing showed that JL94 complete gene 6 cDNA is 1356bp.The sequence was analyzed homologously in comparison with the VP6 nucleotide sequences of two reference porcine rotavirus OSU(subgroup Ⅰ)and Gottfried(subgroup Ⅱ).Nucleotide sequence comparison indicated that JL94 shared 86.85% and 82.41% identities with OSU and Gottfried respectively.The deduced animo acid sequences were 97.48% and 93.20% correspondingly.The result showed that JL94 isolate is more similar to OSU in gene type.更多还原
