脑梗死纤维蛋白原β-148 C/T基因多态性与血浆纤维蛋白原功能和水平的关系

【摘要】 目的 探讨脑梗死 (CI)患者纤维蛋白原 (Fg) β - 14 8C/T基因多态性频率分布及其与Fg功能和浓度的关系。 方法 用聚合酶链反应 限制性片段长度多态性 (PCR RFLP)分析方法 ,对CI组 (15 1例 )和健康对照组 (113名 )进行研究 ,应用Fg功能自动化检测系统检测血浆Fg分子功能参数 ,用比浊法测Fg水平。结果 CI组T等位基因的频率为 0 32 1,对照组为 0 2 4 8(P <0 0 5 ) ;CI组和对照组Fg分子功能 (FMPV/Amax)分别是 4 4 2 9± 0 5 0 9和 3 6 87± 0 4 35 (P <0 0 1) ,Fg浓度(g/L)分别为 3 97± 0 37和 2 97± 0 5 1(P <0 0 1)。女性患者Fg分子功能则比对照组明显增高 (P<0 0 1)。结论 Fg分子功能增强和Fg浓度升高是CI的危险因素 ,Fgβ - 14 8C/T基因多态性与血浆Fg分子功能和Fg浓度具有相关性 ;Fg在脑梗死发生的机制中男性与女性可能有所不同。

【Abstract】 Objective To investigate the frequency of beta-fibrinogen(Fg) gene-148 C/T polymorphism and its association with plasma fibrinogen function and level in patients with cerebral infarction(CI). Methods The beta-Fg gene-148C/T polymorphism of 151 patients with CI and 113 health individual controls were analyzed by restriction fragment length polymorphism(RFLP).Fg molecular reactivity was measured by kinetic method system, and Fg level was determined using turbidimetry. Results The frequencies of T allele in CI patients and health controls were 0.321 and 0.248,respectively( P <0.05).The plasma Fg molecular reactivity was enhanced in cases of CI than in health controls, 4.429±0.509 and 3.687±0.435 ,respectively ( P <0.01). The plasma Fg level of CI patients was significantly higher than that of the health controls, 3.97±0.37 and 2.97±0.51( P <0.01),respectively. In female patients Fg molecular reactivity was higher than in controls( P <0.01). Conclusions The increased plasma Fg level and enhanced Fg molecular are both risk factors of CI. The beta Fg gene-148 C/T polymorphism should be correlated to the plasma Fg molecular reactivity and Fg level. It suggests that the mechanism linking Fg and the development of CI should be different in males and females.
