【摘要】 目的　探讨对氧磷酯酶 192Gln/Arg基因多态性与 2型糖尿病合并冠心病的关系。　方法　采用多聚酶链反应 限制性片段长度多态性 (PCR RFLP)法检测 10 4例健康对照者 (作为正常对照组 )和 80例 2型糖尿病、96例 2型糖尿病合并冠心病 (CDH)患者PON1 192位点的多态基因型。测量体重指数、总胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白、载脂蛋白AI及载脂蛋白B。　结果　 ( 1)中国汉族人PON1 192位点QQ、QR、RR基因型频率及Q、R等位基因频率 :2型糖尿病合并CHD组与正常对照组比较差别有显著性 ;与单纯糖尿病组比较差别无显著性。 ( 2 )中国人与白种人PON1 192位点基因型频率及等位基因频率比较差别非常显著。 ( 3)含R等位基因的基因型QR、RR是 2型糖尿病合并CHD的独立危险因素。　结论　对氧磷酯酶 192Gln/Arg基因多态性与 2型糖尿病合并CHD相关。更多还原

【Abstract】 Objective To investigate the association of paraoxonase 192Gln/Arg gene polymorphism with coronary heart disease in Chinese Han type 2 diabetes mellitus. Methods Paraoxonase 192Gln/Arg gene polymorphism was determined in 280 individuals of 3 groups, matched by age and gender, including 80 patients with type 2 diabetes mellitus, 96 type 2 diabetic patients with coronary heart disease and 104 healthy subjects as control. The paraoxonase 192Gln/Arg genotype was assessed using polymerase chain reaction and restriction fragment length polymorphism (PCR RFLP) method. Variables, including BMI, TC, TG, HDL, LDL, ApoAI, ApoB and paraoxonase 192Gln/Arg genotype, were analyzed by multivariate logistec regression to determine the independent risk factors for coronary heart disease in type 2 diabetes mellitus. Results The frequency of the paraoxonase 192Gln/Arg genotype and the frenquency of the R allele showed significant difference between type 2 diabetic patients with coronary heart disease and control subjects (P<0.05), but there was no difference in genotype frequencies or allele frequencies between type 2 diabetic patients with and without coronary heart disease( P >0.05). The most frequent genotype and allele of 192 Gln/Arg polymorphism of paraoxonase gene in Chinese Han were genotype QR and allele R, but were genotype QQ and allele Q in the White. Their frequency distribution showed significant difference between Chinese and the White. The genotypes carrying R allele were independent risk factors in diabetic patients with coronary heart disease. Conclusion Paraoxonase 192Gln/Arg gene polymorphism is associated with coronary heart disease in Chinese Han type 2 diabetes mellitus.更多还原
