中枢白细胞介素-1β对大鼠全脑缺血后学习记忆的影响
The effects of central interleukin-1β on memory and learning in global cerebral ischemia in rat
【摘要】 目的:探讨白细胞介素-1β(IL-1茁)对全脑缺血再灌注大鼠学习记忆的影响。方法:脑室注射(icv)IL-1茁及抗体后,采用四动脉结扎法制备全脑缺血再灌大鼠模型。于全脑缺血10min后再灌注的6—9d,利用Morris水迷宫,测试大鼠学习记忆功能,并对大鼠海马CA1区正常神经进行计数。结果:①给IL-1β预处理的缺血大鼠记忆功能明显下降,其在Morris水迷宫内的逃逸潜伏期、游泳距离延长,但游泳速度却无改变;②各组比较,学习功能未发现显著性变化;③给IL-1茁预处理的缺血大鼠,海马CA1区损伤神经百分数明显高于对照组。结论:IL-1茁加重了脑缺血再灌注大鼠记忆障碍,其作用可能与海马CA1区神经元严重损伤有关。
【Abstract】 Objective:To investigate the roles of interleukin-1茁(IL-1茁)on changes of rat memory and learning af鄄ter global cerebral ischemia.Method:After intracerebroventricular injection(ICV)of IL-1茁or its antibody,rats under鄄went transient global cerebral ischemia using four-vessel occlusion.The ability of memory and learning was studied by water-maze on6-9days test after10min global cerebral ischemia.The number of normal neurons were counted.Result:①ICV IL-1茁before global cerebral ischemia exaggerated memory impairment,it caused the es鄄cape latency,path length prolong while there were no significant differences in swimming speed;②Learning im鄄pairment was not significantly affected by ICV IL-1茁or its antibody;③The percentage of damaged neurons of hipocamus CA1areas in ICV IL-1茁and global cerebral ischemia group was significantly higher than those of the control group.Conclusion:Central IL-1茁enlarges memory impairment after global cerebral ischemia in rat,IL-1茁is likely due to the exacerbated neuronal damage in hipocamus CA1areas.Author ’s address Dept.of Physiology and Pathophysiology,Dali Medical College,Yunnan,671000
