甲基苯丙胺对大鼠心肌线粒体抗氧化酶的影响
Effect of methamphetamine on antioxidative enzymes and isozymes of cardiac mitochondria of mice
【摘要】 目的 甲基苯丙胺对大鼠心肌线粒体抗氧化酶的影响。方法 采用不同浓度的甲基苯丙胺 (1 0、1 5,2 5和 4 0mg/kg)通过体内染毒方式作用于大鼠 ,观察其心肌线粒体谷胱甘肽过氧化物酶 (GSH -Px)和超氧化歧化酶 (SOD)活性的变化 ,并使用聚丙烯酰胺凝胶电泳法进一步分析这 2种酶的同工酶谱。结果 低浓度的甲基苯丙胺对这两种酶的活性有一定的激活作用 ,而高浓度的甲基苯丙胺则有着明显的抑制作用 ,同工酶谱中出现了特异性标志酶sod1,4,5和gsh4。结论 甲基苯丙胺能导致大鼠心肌线粒体产生大量的活性氧自由基 (ROS) ,并进一步对线粒体的结构和功能产生破坏性的影响 ,包括对线粒体DNA及其转录系统的作用等。这可能是甲基苯丙胺心肌毒性的主要作用机制之一。
【Abstract】 Objective To explore effects of methamphetamine(METH)on a ntioxidative enzymes of cardiac mitochondria of mice.Methods The effect of different concentration (1.0,1.5,2. 5 and 4.0?mg/kg)METH on the activities of glutathione peroxidase(GSH-Px) and su peroxidative dismutase(SOD) of cardiac mitochondria of mice through belly inject ion were discussed,and the patterns of the two isozymes(GSH-Px and SOD) were att ained using PAGE method.Results Low concentration METH can activate GSH-Px and SO D while the high concentration METH can restrain them,and some symbol isozym es sod1,4,5 and gsh4 were found in the patterns of isozymes.Conclusion The administration of METH on mice in vivo can lead to excessive active oxygen radicals in cardiac mitochondria which further injured mitochondrial constitute parts,including DNA and its encoding system,thi s maybe mainly toxicol mechanism of METH on mcie cardiac.
