【摘要】 转化生长因子TGF β/Smads是细胞增殖的抑制因子。它可以促进细胞外基质的增生和原发性肿瘤的转移。选择 2 0例肺癌组织标本及 8种不同转移能力的肺癌细胞系 ,通过荧光免疫组织化学染色法进行RⅡ和Smads蛋白定位及表达分析。初步探讨了TGF β/Smads信号传导通路与肺癌发生和转移的关系。

【Abstract】 Smad proteins transduce signals from transforming growth factor beta superfamily ligands that regulate cell proliferation,differention and death through activation of receptor serine/threonine kinases.TGF β/Smads signal pathway not only has transforming potential but can also drive tumourigenesis,malignant progression,invasion and metastasis of human cancers.Using the immunohistochemistry,we investigate the expression and location of TGF βRⅡ,Smad2,Smad4 and Smad7 in 20 lung cancer specimens and 8 lung cancer cell lines.The results suggest that aberrant smads protein expression is significantly related to lung cancer tumoruigenesis and progression.Interestingly,TGF βRⅡ and Smad7 strongly express in high metastasis cell lines.High expression of TGF βRⅡ and smad7 in the cell lines with high metastatic potential showed a conceivable TGF β signal pathway independent Smads in the lung cancer,and that might mediate invasion and metastasis of lung cancer.
