【摘要】 目的 研究甘油三酯 (triglycerides,TG)对胰岛细胞的作用 ,并探讨NO 1886对TG诱导的胰岛细胞凋亡的作用和对胰岛 β细胞功能的作用。 方法 用TG处理原代培养的胰岛 ,MTT法检测细胞活性 ,原位凋亡法及ELISA法检测细胞凋亡 ,放射免疫法检测胰岛素的含量。结果 TG对细胞生长呈浓度及时间依赖性抑制作用。原位凋亡法及ELISA法证实TG可诱导胰岛细胞凋亡 ,而NO 1886可抑制TG诱导的胰岛细胞凋亡 ;放射免疫法结果显示NO 1886处理的胰岛分泌的胰岛素与对照组相比增加 (P <0 0 5 )。结论 TG对胰岛的细胞毒作用与其作用剂量及作用时间有关 ;高浓度的TG可能通过致胰岛细胞凋亡的途径使胰岛素分泌减少 ;NO 1886可对TG诱导的胰岛细胞凋亡起保护作用 ,其本身具有刺激胰岛β细胞分泌胰岛素的作用

【Abstract】 AIM To elucidate the effect of triglyceride on the cultured islet cells and study the effect of NO 1886 on triglyceride induced islet cells apoptosis and the function of islet cells. METHODS MTT test was carried out to evaluate the islet cells growing state with different concentrations and time of triglyceride incubated with islet cells; Terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) test was conducted to analyze apoptosis; ELISA was conducted to analyze apoptosis index and radioimmunoassay was carried out to evaluate insulin. RESULTS The inhibiting effect of TG on the islet cells growth by TG was in a dose and time dependent manner. TUNEL test showed that triglyceride induced islet cells apoptosis and ELISA test showed the apoptosis index of NO 1886 group was decreased compared with TG group( P< 0 05); radioimmunoassay showed the insulin values of NO 1886 groups were significantly increased compared with the control groups ( P <0 05). CONCLUSION The cytoxoicity of triglyceride to islet cells is dosage and time dependent; the triglyceride can inhibit the cultured islet cell growth, and even result in cell apoptosis; NO 1886 can protect islet cells from triglyceride induced apoptosis and stimulate insulin secretion from the pancreatic β cells.
