人nov基因全长cDNA克隆、测序及其组织表达谱分析

【摘要】 采用特异性mRNA富集技术,从人早期胚胎中得到人nov基因(novH)的cDNA克隆,序列分析表明,该基因cDNA序列由2389个碱基对组成,包含一段长1071bp的开放阅读框(ORF),3′端含有加尾信号AATAAA和ply(A)尾巴.Northern杂交显示,在Hela细胞和肾上腺组织中有表达的novHmRNA,大小约为2.5kb;多组织RNA点杂交分析结果进一步表明,novH基因在肾上腺和主动脉中的表达水平相对较高,在成人肾、肝、肺和小肠组织中亦有少量的表达.

【Abstract】 The nov gene encodes a cysteine-rich protein that is overexpressed in avian nephroblastomas. A full-length cDNA encoding human nov gene was isolated from nov-enriched mRNA of early human embryo. The complete cDNA sequence of nov contained an open reading frame (ORF) of 1 071 necleotides and a tailing signal of AATAAA and poly (A) in 3′-end region. Northern blot analysis showed that nov mRNA was mainly expressed in HeLa cell and adrenal gland at 2.5 kb in size closing to the length of the cDNA clone. RNA Master Blots with 50 different RNA from human fetus and adult tissues was used to further detection of novH gene. The results showed that novH gene was expressed at relatively high level in normal human adrenal gland and aorta. The expression of novH gene was also detected at lesser extent in kidney, live, lung and small intestine. This suggests that nov gene may play an important part not only in development of the tumors but also in the normal adrenal gland and aorta.
