诱导型与内皮型一氧化氮合酶在小鼠输卵管黏膜中的表达与活性

【摘要】 目的:探讨诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)与内皮型一氧化氮合酶(endothelial nitric oxide synthase,eNOS)在动情周期小鼠输卵管黏膜的表达规律与活性。方法:间接免疫组织化学法结合计算机图象分析技术。结果:iNOS和eNOS在动情周期小鼠输卵管黏膜内的表达模式(expression pattern)非常相似。在间情期,黏膜中iNOS和eNOS阳性细胞较少,强度较弱;从动情前期到动情后期,iNOS和eNOS阳性细胞数量明显增多,强度也逐渐提高,并在动情后期达顶峰。与此相一致,3-硝基酪氨酸(3-nitrotyrosine,3-NT)在黏膜上皮中的表达丰度也以发情后期最高。结论:iNOS和eNOS在生育期小鼠输卵管黏膜的表达具有明显的周期性,动情后期表达高活性的iNOS和eNOS,可能是输卵管作为受精部位的原因之一。

【Abstract】 Objective:To investigate the expression patterns and enzymatic activity of both inducible nitric oxide synthase (iNOS) and endothelial nitric oxide synthase (eNOS) in cycling oviduct. Methods :Indirect im-munohistochemical protocol was employed for this study with assistance of computer imaging analysis system. Results:An extremely similar expression pattern of both iNOS and eNOS was detected in the oviducts during the estrous cycle. In dioestrus , a weaker positive reaction showed at the epithelial cells of most villi while a few of positively stained cells localized at the basal segments of villi. The number of both iNOS and eNOS positive cells and their intensity in mucosa increased gradually during the cycle. However, the highest intensity of iNOS and/or eNOS in immunohistochemical reaction appeared in metoestrus. The detection of 3-NT in cycling oviduct showed the same pattern with that of NOSs. Conclusion: This paper provides the evidence that both the expression patterns and enzymatic activity of iNOS and eNOS in oviduct are significantly characteristic of estrous cycle in mice. The highest intensity of immunohistochemical reaction and the highest activity of NOSs in metoestrous oviduct may indicate that oviduct functions in such a way would be one of the natural reasons why fertilization always takes place at oviduct in mammals.
