磷脂酰胆碱特异性磷脂酶C在人肺癌A549细胞分化中的作用研究

【摘要】 为探讨磷脂酰胆碱特异性磷脂酶C在人肺癌肺泡Ⅱ型上皮细胞A5 4 9分化中所起的作用 ,本文利用该酶的特异性抑制剂D6 0 9抑制此酶的活性 ,用倒置相差显微镜观察了D6 0 9处理体外培养的人肺癌A5 4 9细胞形态变化 ,用流式细胞技术测定了D6 0 9对A5 4 9细胞周期及凋亡率的影响 .结果发现D6 0 9促使该细胞拉长 ,并向Ⅰ型上皮细胞分化 ,表现出形成肺泡结构的趋势 ,同时改变了细胞周期的分布 :G0 1 期细胞数目增加 ,S期和G2 M期细胞数目明显减少 .结果表明 ,磷脂酰胆碱特异性磷脂酶C在使A5 4 9细胞维持其Ⅱ型细胞状态中具有重要作用 .

【Abstract】 To explore the role of phosphatidylcholine specific phospholipase C in A549 lung carcinoma cell differentiation, when the activity of phosphatidylcholine specific phospholipase C was suppressed by D609, a specific inhibitor of this enzyme, the morphological changes of cells were observed with inverted microscope, and the cell cycle distribution and the percent of apoptotic cells were examined with flow cytometry. We found that the cells became elongated and had the trend of forming pulmonary alveoli. At the same time, the cell cycle distribution changed, the cells in phase of G 0/1 increased and those in S and G 2 /?M phase decreased significantly. These results showed that phosphatidylcholine specific phospholipase C might play an important role in keeping the type Ⅱcell state of A549.
