中国人高血压、冠心病及糖尿病者的血管紧张素原基因Thr 174Met及Met 235Thr变异分析

【摘要】 目的了解血管紧张素原(AGT)基因是否参与中国人高血压(HTN)、冠心病(CHD)及2型糖尿病(T2DM)发病。方法358例呈HTN、CHD及T2DM不同组合存在的中国人。检测AGT基因外显子2Thr174Met(T174M)及Met235Thr(M235T)两个变异,用单变异及多变异统计方法分析其疾病相关情况。结果AGT基因与中国人HTN相关。主要表现在T174M变异上。在HTN单一疾病组、HTN合并CHD及/或T2DM组、HTN总组以及HTN年龄亚组均见基因型频率分布与对照组间有显著差异(P=0.024~0.049)。HTN者174T纯合子频率(P=0.019~0.030)及174T等位基因频率增高(P=0.045~0.052)。逐步回归分析表明AGT-T174M是HTN的独立相关因素。T174M与M235T间存在明显连锁不平衡。AGT基因与CHD及T2DM无相关。结论AGT基因是参与中国人HTN的遗传因素之一。

【Abstract】 Objecive To ascertain whether angiotensinog en gene(AGT)contribute to the development of hyp ertension(HTN),coronary heart disease(CHD)and type 2diabetes mellitus(T 2 DM)in Chinese.Methods 358Chinese subjects with various combination of HTN?CHD and T 2 DM were included in this study.Two va riants of the exon 2of AGT -Thr174Met(T174M)and Met 235Thr (M235T)were genotyped.The association of t hese variants with HTN?CHD and T 2 DM was ana-lyzed with univariate and multi -variate statistic analysis.Results AGT was associated with HTN in Chinese.T175M was the more prominent one.The genotype fre quency distributions showed signif icantly difference between groups o f HTN(simple,combined with CHD and /or T 2 DM total)or age -subgroup of HTN and controls(P=0.024~0.049),with increase in fre-quencies of 174T homozygotes(P=0.019~0.030)and 174T allele(P=0.045~0.052)in subjects with HTN.Stepwise regression analysis showed that AGT-T174M was an independent related fa ctor for HTN.Significant linkage di sequilibrium was observed between T174M and M235T.AGT was not associated with CHD and T 2 DM.Conclusions AGT in one of the genetic factors responsible for the development of HTN in Chinese.[
