重肌灵对EAMG大鼠免疫调节作用的实验研究
Experimental Study of the Immunoregulatory Effects of Zhongjiling in the Ratswith Experimental Autoimmune Myasthenia Gravis
【摘要】 目的 研究中药复方重肌灵对实验性自身免疫性重症肌无力 (EAMG)大鼠免疫调节作用。方法 建立Lewis大鼠EAMG模型 ,将动物分为正常组、佐剂组、模型组、重肌灵组。分别采用 3H TdR掺入法、ELISA法 ,从N2 AchR对淋巴结细胞特异性增殖反应、血清中AchRab滴度、血清IL 4、IFN γ水平等方面观察重肌灵对EAMG大鼠免疫功能的调节作用。结果 与模型组相比 ,重肌灵组可使N2 AchR所致的淋巴结细胞增殖反应受到抑制 (P <0 0 1) ;血清AchRab滴度明显降低 (P <0 0 1) ;血清IL 4、IFN γ水平下调 (P <0 0 1)。结论 重肌灵可能是通过下调血清IL 4、IFN γ ,抑制淋巴结细胞特异性增殖反应 ,抑制AchRab产生而起到治疗作用
【Abstract】 Objective To study the immunoregulatory effects of Zhongjiling (ZJL), a TCM compound preparation, in the rats with experimental autoimmune myasthenia gravis (EAMG). Methods Lewis rat was used to establish the EAMG model. There were 4 rat groups in the experiment, namely the normal control, CFA, model, and ZJL. The immunoregulatory effects of ZJL on EAMG were observed through detecting the changes in the N 2AchR-induced specific proliferation reaction of the lymphocytes in lymph nodes, serum AchRab titer, and the levels of serum IL-4 and IFN-γby JH3,D,Z-TdR permeation method and ELISA. Results In the ZJL-group, as compared with the model group, the N 2AchR-induced specific proliferation reaction of the lymphocytes in lymph nodes was inhibited (P<0.01), the serum AchRab titer was lowered (P<0.01), and the levels of serum IL-4 and IFN-γ were downregulated (P<0.01). Conclusion The mechanism of the immunoregulatory effects of ZJL may be downregulating the levels of serum IL-4 and IFN-γ, inhibiting the specific proliferation reaction of the lymphocytes in lymph nodes, and restraining the production of AchRab.
