修复基因hMSH2在肺癌发生中的作用

【摘要】 目的 探讨错配修复基因 h MSH2 在人群肺癌发病中的作用。方法  RT- PCR技术检测细胞中 h MSH2 基因的 m RNA表达 ;用免疫组化检测基因的蛋白表达。结果  32 .0 % ( 4 8/ 15 0 )的肺癌组织和 5 .0 % ( 2 / 4 0 )的正常肺组织存在 h MSH2 基因表达低下 ,修复基因 h MSH2 表达低下与肺癌发生有关联 ,OR值为 8.94 ( 1.99,5 5 .94 ) ,P <0 .0 1,估计其人群归因危险度 PAR%为 7.71%。16 .0 % ( 8/ 5 0 )的肺癌病人外周血和 2 .0 % ( 1/ 5 0 )的正常人外周血单个核细胞也可检出 h MSH2 基因表达低下。分析各种暴露因素和临床特征对 h MSH2 基因表达的影响 ,未发现与该基因表达相关的因素。但发现 ,p5 3突变蛋白的过度表达与 h MSH2 表达低下有一定的关联 ( P <0 .0 5 )。结论 DNA修复基因 h MSH2 在肺癌发病中起着重要的作用 ,该基因表达低下可作为有价值的肺癌易感性标志

【Abstract】 Objective To investigate the role of DNA mismatch repair gene hMSH 2 in the development of human lung cancer. Methods The reverse transcription polymerase chain reaction (RT PCR) method was applied to measure hMSH 2 mRNA expression. The protein expressions of genes were assessed by immmuno histochemistry. Results The expression of hMSH 2 was low or absent in 48 of 150 (32.0%) lung cancer specimens, whereas only in 2 of 40 (5.0%) normal lung tissues reduced levels of hMSH 2 expression. Low expression of DNA repair gene hMSH 2 was associated with the development of human lung cancer, OR was 8.94 (1.99, 55.94), P<0.01, the population attributable risk proportion (PAR%) was 7.71%. Reduced expression levels of hMSH 2 were observed in 8 of 50 (16.0%) lung cancer patients peripheral blood mononuclear cells, and 1 of 50 (2.0%) normal controls blood cells. In the investigation of the exposure factors and clinical features that may affect the expression of hMSH 2 gene, but no factor showed correlation with hMSH 2 . The over expression of p53 was correlated with the low expression of hMSH 2 , P< 0.05 . Conclusions DNA repair gene hMSH 2 may play an important role in the development of human lung cancer, and the low expression of hMSH 2 could be used as a valuable susceptibility biomarker of human lung cancer.
