【摘要】 目的 :探讨卵巢颗粒细胞中凋亡相关蛋白bax的表达与妊娠率的关系。方法 :选择 4 2例适合作ICSI手术的不孕妇女 ,其中由于男性少精及无精的 2 1例 ,男性免疫性不孕 6例 ,女性免疫性不孕 7例 ,男女双方免疫性不孕 8例。根据妊娠情况 ,将其分为妊娠组 (18例 )、非妊娠组 (2 4例 )。通过脱氧核糖核苷酸末端转移酶介导的缺口末端标记法 (TUNEL)和免疫组织化学方法检测卵巢颗粒细胞凋亡率和bax蛋白表达率。结果 :妊娠组卵巢颗粒细胞凋亡率为 0 .17± 0 .0 2 ,显著低于非妊娠组 0 .36± 0 .0 3,P <0 .0 5。妊娠组bax阳性率为 0 .33± 0 .0 2显著低于非妊娠组 0 .6 6± 0 .0 4 ,P <0 .0 5。结论 :卵巢内卵丘颗粒细胞凋亡状态和凋亡相关蛋白bax的表达与胚胎质量有直接的关系 ,研究卵丘颗粒细胞凋亡和卵丘颗粒细胞的bax基因表达可作为预测ICSI成功的指标更多还原

【Abstract】 Objective: To investigate the relation between the incidence of apoptosis and expression of apoptosis related protein bax in human cumulus granulosa cells(GCs) and pregnancy rate after intracytoplasmic sperm injection (ICSI). Mothods: 42 cases women undergoing ICSI were retrospectively analysed, 21 cases with oligasthenoteratosperm,6 cases with male immue infertility,7 cases with female immune infertility and 8 cases with couples immune infertility. The incidence of apoptosis and the expression of bax protein in cumulus GCs were detected by terminal deoxynucleotidyl transferase mediated dUTp nick end labing (TUNEL) and immunohistochemistry method respectively. Results: apoptosis of cumulus GCs in pregnant group was significantly lower than that in non pregnant group (0.17±0.02) vs(0.36±0.03), P<0.05. The expression of bax protein in cumclus GCs were significantly lower than that in non pregnant group(0.33±0.02)vs(0.66±0.04). Conclusion: The data indicates that women with pregnancy after fresh ICSI have significant lower incidence of apoptosis and expression of bax protein in cumulus GCs compared with those without pregnancy.更多还原
