三倍体毛白杨组培快繁技术

【摘要】 试验研究了不同基本培养基(1/2MS和WPM)和不同浓度的NAA和BA组合对三倍体毛白杨离体快繁的影响.结果表明:1/2MS+BA0.3mg·L-1+NAA0.05mg·L-1为适宜的增殖培养基,芽分化率达86.4%,平均芽数达4.4;1/2MS+NAA0.05mg·L-1为适宜的壮苗生根培养基,生根率达94.1%,平均根数为3.1,平均根长为3.5cm,试管苗平均高度达7.4cm;在移栽过程中发现,带愈伤组织的试管苗成活率高于不带愈伤组织的试管苗,达97.0%.

【Abstract】 This test studied the influences of different primary mediums (1/2MS and WPM) and different concentration of NAA and BA on the tissue culture rapid propagation of triploid populus tomentosaThe results were summarized as follows:1/2MS+BA03 mg·L-1+NAA005 mg·L-1 was the optimun medium,the sprouting bud rate arrived at 864% and the average number of buds was 44;1/2MS+NAA005 mg·L-1 was the optimun strengthening seedling and rooting medium,whose rate of rooting arrived at 941%,the average number of roots was 31,the average length of the roots was 35 cm and the average height of the seedling from test tubeswas 74 cm;the percent of live plant of the seedlings from test tubes with callus was 970% which was higher than those without callus,in the course of transplanting
