雄黄对K562细胞端粒酶活性和凋亡的作用
Effects of realgar on telomerase activity and cell apoptosis in K562 cells
【摘要】 目的 :探讨雄黄对慢性粒细胞白血病细胞株K5 6 2细胞端粒酶活性及凋亡的调节作用和机制 .方法 :MTT法检测细胞增殖力 ;PCR ELISA法测定端粒酶活性 ;流式细胞仪测量细胞周期和细胞凋亡 .结果 :0 .3,0 .6 ,1.5和 3.0mg·L-1雄黄 (72h)在诱导K5 6 2细胞凋亡的同时可显著抑制细胞端粒酶活性和细胞增殖 ,并呈剂量相关 ;1.5 ,3.0mg·L-1雄黄 (72h)可诱导K5 6 2细胞G2 /M期阻滞 .结论 :雄黄诱导K5 6 2细胞凋亡达到抑制细胞生长的作用 ,端粒酶活性下降是雄黄诱导K5 6 2细胞凋亡的机制之一 .由大剂量雄黄诱导的K5 6 2细胞G2 /M期阻滞可能与端粒酶活性显著下降有关
【Abstract】 AIM: To explore the effects of realgar on telemorase activity and cell apoptosis in K562 cell line. METHODS: Cell proliferation was analyzed by MTT assay and telomerase activity was determined by polymerase chain reaction enzyme linked immunoassay (PCR ELISA). Flow cytometry was applied to analyze the cell cycle and apoptosis. RESULTS: 0.3, 0.6, 1.5 and 3.0 mg·L -1 realgar (72 h) significantly induced apoptosis of K562 cells following the remarkable inhibition of telomerase activity and proliferation in K562 cells. G2/M phase arrest appeared following treatment with realgar in 1.5 and 3.0 mg·L -1 (72 h). CONCLUSION: Realgar inhibits the cell growth by inducing apoptosis of K562 cells and the down regulation of telomerase activity perhaps is one of the mechanisim of realgar induced apoptosis in K562 cells. The G2/M phase arrest in K562 cells induced by large dose of realgar might be related to the significant reduction of telomerase activity.
