芪麝颈康丸调控大鼠颈椎间盘碱性成纤维细胞生长因子及胰岛素样生长因子mRNA表达的研究

【摘要】 目的:探讨芪麝颈康丸对大鼠退变颈椎间盘碱性成纤维细胞生长因子(bFGF)及胰岛素样生长因子(IGF-Ⅰ)mRNA 表达的调节作用。方法:建立动静力失衡性颈椎间盘退变模型,逆转录多聚酶链式反应方法检测椎间盘组织内 bFGF、IGF-ImRNA 表达水平。结果:模型组 bFGF mRNA 表达明显,与正常对照组比较差异有显著性(P<0.01);与模型组比较,芪麝颈康丸、活血通络方、芬必得都可以下调 bFGF,尤以芪麝颈康丸下调明显(P<0.01)。模型组 IGF-Ⅰ mRNA 表达明显减弱,与正常对照组比较差异有显著性(P<0.01);芪麝颈康丸可以上调 IGF-ⅠmRNA(P<0.01),活血通络方、芬必得作用不明显。结论:芪麝颈康丸可以在下调椎间盘组织内 bFGF mRNA,上调 IGF-Ⅰ mRNA 的基因表达。

【Abstract】 Objective:To study the effects of Qishe Jingkang Pills(QSJKP)on the expression of basic fibrocyte growth factor(bFGF)and insulin-like growth factor-I(IGF-I)mRNA in the degenerated intervertebral disc.Methods:The model of rats degeneration of cervical intervertebral discs caused by imbalance between the dynamic and static force was established.The gene expression of bFGF and IGF-1 mRNA in degeneration cervical intervertebral disc was assayed by reverse transcription polymerase chain reaction(RT-RCR).Results: The expression of bFGF mRNA in the intervertebral disc in model group was markedly higher than that in normal group,and some sig- nificant difference was shown in comparing with QSJKP.The expression of IGF-I mRNA in the intervertebral disc in model group was obviously lower than that in normal group,and significant difference was shown in comparing with QSJKP and model group.Conclu- sion:QSJKP may use to down-regulation gene expressed of bFGF and up-regulation of IGF-I mRNA.
