脾虚大鼠延髓网状结构小泡乙酰胆碱转运体免疫组化研究
Immunohistochemical Study on Vesicular Acetylcholine Transporter of Cholinergic Neurons in the Reticular Formation of Medulla Oblongata in Spleen Deficiency Rats
【摘要】 目的 :探讨中医脾虚证与胆碱能神经功能之间的关系。方法 :选用 30只 Wister大鼠用利血平制作中医脾虚动物模型 (利血平组 ) ,并选用特异性较强的胆碱能神经递质乙酰胆碱标志物——小泡乙酰胆碱转运体(VACh T)对延髓网状结构进行免疫组化研究。并与 30只用 0 .9% Na Cl溶液大鼠 (对照组 )对照。结果 :利血平组大鼠 VACh T免疫反应与对照组相比明显增强。细胞胞体上及边缘突起上 ,颗粒数目 2 0~ 4 0个 /每个神经细胞内 ,颗粒染色明显加深 ,颗粒较大 ,排列紧密。结论 :这种内源性乙酰胆碱的变化考虑为中医脾虚的本质所在。
【Abstract】 Objective: To study the relationship between spleen deficiency syndrome of TCM and the function of cholinergic neurons. Methods:Thirty Wister rats’ spleen deficiency model was made with Phenethyldinuanide,and vesicular acetylcholine transporter (VAChT), a very peculiar marker of cholinergic nervous delivers, was selected for the immunohistochemical study of cholinergic neurons in the reticular formation of medulla oblongata. Thirty rats in the control group were disposed with 0.9% NaCl solution.Results: VAChT immunohistochemical reaction was obviously strengthened in Phenethyldiguanide-treated group as compared with that in the control group.Conclusion: The internal change of acetylcholine was considered as the essence of spleen deficiency of TCM.
