酒精性肝病大鼠Kupffer细胞CD14蛋白的表达及其在肝损害中的作用

【摘要】 目的:观察大鼠酒精性肝病时细胞()蛋白和基因的表达及其在肝损害中的作用。KupfferKCsCD14方法:随机将大鼠分为乙醇喂养组(剂量~)和葡萄糖对照组。周和周活杀分离,用流式细胞仪()测定细Wistar 512g/kg/24h48KCsFCM胞膜上蛋白的表达,同时用逆转录多聚酶联反应()测定中的表达;用酶联免疫法()CD14RT-PCRKCsCD14 mRNAELISA测定血浆中TNF-α和-浓度变化;测定血浆内毒素及血浆中转氨酶()水平变化,并观察肝脏形态学改变。IL6ALT结果: 乙醇组大鼠周时膜上已明显表达蛋白和 ,周时其表达进一步增强,显著高于对照组4KCsCD14CD14mRNA8(P。乙醇<0.05)组周和周时血浆48TNF-α浓度、-浓度及内毒素浓度均明显高于对照组IL6 (P。乙醇组肝组织发生显著的病理变化,对<0.05)照组肝组织无明显病理变化。结论:乙醇能诱导大鼠肝脏膜蛋白和的表达显著增强,血浆中细胞因子KCsCD14CD14 mRNA-TNFα和-浓度增加,引起肝脏形态改变和功能损害。IL6

【Abstract】 Objective: To observe the expression of CD14 protein and its mRNA on Kupffer cells (KCs) and evaluate the role of CD14 protein in the pathogenesis of liver injury in the rats with alcohol-induced liver disease (ALD). Methods: Twenty eight Wistar rats were divided into two groups: ethanol-fed group and control group. Ethanol-fed group were fed ethanol(dose of 5~12/kg/24h) and control group received dextrose instead of ethanol. Two groups were sacrificed at 4w and 8w. KCs were isolated and expression of CD14 protein and its mRNA on KCs were determined with flow cytometric (FCM) analysis or the reverse transcription polymerase chain reaction (RT-PCR) analysis. Levels of tumor necrosis factor (TNF)-α and interleukin(IL)-6 in plasma were determined with enzyme linked immunosorbent assay (ELISA). Levels of endotoxin and alanine transaminase(ALT) in blood were measured also, and liver pathology was observed under light and electron microscopy. Results: In ethanol-fed rats, ethanol administration led to a significant expression in CD14 protein and its mRNA compared with the control group(P<0.05) at 8 week. In ethanol group, concentrations of TNF-α, IL-6 and plasma endotoxin levels were increased dramatically than in the control rats at two time points. In liver section from ethanol-fed rats, there was marked pathological changes but there was no change in control group. Conclusion:In ethanol-fed rats, ethanol administration led to a significant expression of CD14 protein and its mRNA, and increase of TNF-α and IL-6 levels, which may result in the pathological changes of liver tissue and hepatic functional damages.
