溴氰菊酯抗性家蝇酯酶的生化分析

【摘要】 目的 :研究拟除虫菊酯对家蝇的抗性机制。方法 :通过对溴氰菊酯敏感、抗性和高抗性家蝇的离体酯酶活力测定、酶反应动力学及不连续非变性凝胶电泳 ,观察酯酶在拟除虫菊酯抗性中的作用。结果 :用α -NA做底物酯酶活力 ,Vm 在 3种品系家蝇中无差异 ,2个抗性品系之间Km 和Vm 也无差异 ,但敏感品系的Km 分别高于两抗性品系 ;聚丙烯酰胺凝胶电泳表明其同功酶谱差异很大 ,SS有 6条带 ,R、RR各有 5条带 ,且三者之间带的位置和带型均发生改变。结论 :拟除虫菊酯抗性家蝇酯酶性质发生改变 ,可能参与对拟除虫菊酯的代谢抗性

【Abstract】 Objective:To investigate the role of esterase in pyrethroid resistance of housefly.Methods: To detect the activity, kinetic parameters and the polyacryl gel electrophoresis spectrum of the esterase from the susceptible, resistant and super resistant strains. Results: The activity and the V m of the esterase in the three strains were similar. The K m of the esterase from resistant and super resistant strains was also similar. But the K m of the esterase in the susceptible strain was much higher. Further polyacryl gel electrophoresis revealed that the esterase from different strains involves different isoenzyme composition. Conclusion: The changes of esterase isoenzymes is involved in the mechanisms for pyrethroid resistance in housefly.
