【摘要】 目的　检测活动性狼疮性肾炎 (LN)患者外周血和肾组织内浸润T淋巴细胞受体 (TCR)基因亚家族的限制性取用 ,以探讨体内自身反应性T细胞克隆存在的证据。方法　用半定量逆转录聚合酶链反应 (RT PCR)的方法分析 18例活动性LN患者和 12名正常对照者的T细胞受体Vβ亚家族的部分基因表达。结果　活动性LN患者外周血T淋巴细胞受体Vβ8基因表达明显高于正常对照组(0 .6 7± 0 .11vs 0 .36± 0 .0 3,P <0 .0 0 1) ,肾活检组织中浸润淋巴细胞Vβ8基因的表达也明显高于TCRVβ1、TCRVβ11、TCRVβ2 0这些TCRVβ家族的其他成员 (P <0 .0 0 1) ,并呈现出以TCRVβ8为优势克隆的T细胞的大量增殖和聚集。结论　在LN的自身免疫性损伤中 ,存在具同一特征 (TCRVβ8)自身反应性T细胞的克隆聚集、增殖 ,这一发现提示其在LN中可能具有致病作用更多还原

【Abstract】 Objective The limiting usage of T cell receptor (TCR) subfamily genes in T lymphocytes of peripheral blood and intral renal tissue was evaluated in order to verify the existence of autoreactive T lymphocytes clone in patients with active lupus nephritis. Methods With the semi quantitative RT PCR method, the level of TCRVβ subfamily genes expression were analyzed in 18 patients with active lupus nephritis and 12 controls. Results In the patients with active lupus nephritis, expression of TCR Vβ8 gene in lymphocytes of peripheral blood was significantly higher than that of the control (0.67±0.11 vs 0.36±0.03, P <0.001). Expression TCR Vβ8 on infiltrated lymphocytes of renal biopsy tissue was also higher than other subfamilies, i.e. TCR Vβ1, TCR Vβ11, TCR Vβ20 ( P <0.001). Conclusion The results showed that there was a clone gathering and expansion of T lymphocytes with homogenous characteristics (TCR Vβ8+) in the process of autoimmune injury of lupus nephritis, which suggesting the possible pathogenetic effects of autoreactive T cells in patients with lupus nephritis.更多还原
