P物质在再生视网膜节细胞中的表达及IBMX和CPT-cAMP对其表达的影响

【摘要】 采用金黄地鼠视神经切断并缝接坐骨神经的再生实验模型,玻璃体内注射IBMX或/和CPT-cAMP,荧光金逆行标记再生的RGCs结合P物质免疫荧光组化双标法,研究外周神经缝接于视神经断端能否促进P物质阳性的视网膜节细胞(RGCs)再生及IBMX或/和CPT-cAMP处理对其再生的影响。实验结果:①术后四周,对照AG组每个视网膜 再生RGCs数1329±104,双标细胞平均数为45±5,占再生RGCs总数的3.4%;②AG+IBMX组每个视网膜再生RGCs数为2099±419,再生P物质阳性节细胞平均数为119±22,占再生RGCs总数的6.55%;③AG+cAMP组每个视网膜再生RGCs数为2048±133,再生P物质阳性节细胞平均数为127±37,占再生RGCs总数的6.15%;④AG+IB-MX+cAMP组每个视网膜再生RGCs数为4370±487,再生P物质阳性节细胞平均数为339±72,占再生RGCs总数的7.98%,与对照组的差异具有统计学意义。表明成年哺乳动物P物质阳性RGCs能再生,玻璃体内注射IBMX或/和CPT-cAMP可以促进该类RGCs再生。

【Abstract】 We have used the regenerating model of optic nerve(ON) transected and then sutured a segment of autologus sciatic nerve to stump of ON in hamsters, then IBMX or/and CPT-cAMP was injected intravitrally, by using the double-labeled method of Fluro-Gold(FG)retrograde tracing combined with fluorescent immunocytochemistry of substance p(SP); to examine the regeneration of SP-irnrnunoreacttvity (IR)retinal ganglion cells(RGCs)and the effects of 3-isobutyl-l-methylxanthine( IBMX)and/or 8-(4-Chlorophenylthio)-cAMP(CFF-cAMP) injected on regeneration of SPTR RGCs. Our results were:① At 4 weeks after surgery, the mean number of regenerated RGCs per retina in AG group was 1329 ± 104, doubt lable RGCs per retina in AG group was 45 ± 5 which constitutes 3.4 ± 0.33 % of the total regenerated RGCs;② In AG+ IBMX experimental group, the mean number of regenerated RGCs per retina was 2099 ±419,doubt lable RGCs per retina was 119 ± 22, which constitutes 6. 55 ± 3.12 % of the total regenerated RGCs; ③ In AG + CPT-cAMP experimental group, the mean number of regenerated RGCs per retina was 2048 ± 133,doubt lable RGCs per retina was 127 ± 37 which constitutes 6.15 ± 1.10% of the total regenerated RGCs; ④In AG+ IBMX + CPT-cAMP experimental group, the mean numbers of regenerated RGCs per retina were 4370 ± 487, doubt kble RGCs per retina were 339 ± 72, which constitute 7.98 ± 2.21 % of the total regenerated RGCs. The experimental groups were increased significantly compared with those in AG group. The results show that the axotomized SP-IR RGCs have the capability of regenerating their axons into the attached PN graft, It is indicate IBMX and/or CPT-cAMP could enhance the regeneration of the SP-IR RGCs.
