分光光度-白细胞黏附抑制法测定免疫核糖核酸的活性
Determination of the biological activity of immune RNA by spectrophotometry-LAI test
【摘要】 目的建立免疫核糖核酸活性测定方法。方法细胞浓度在 2× 10 6~ 2× 10 7,波长 415nm处吸收值与细胞浓度呈线性关系 ,可用吸收值测出细胞浓度 ,求出免疫核糖核酸的黏附抑制率。结果此法与细胞计数 白细胞黏附抑制 (LAI)实验有良好的相关性 ,r=0 .96 ,P <0 .0 1,重复性实验结果为 :板内RSD值为 6 .3% ,板间RSD值为 5 .4% ,不同实验次数RSD值为 9.4% ,灵敏度 0 .4mg/ml并能反应出免疫核糖核酸的抗原特异性。结论此法特异、简便并具有良好的重复性 ,可以作为免疫核糖核酸的活性测定方法
【Abstract】 PurposeThe aim is to develop a method for determining the biological activity of immune RNA.MethodsUtilization OD value with a linear range of cell consistency 2×10 6~2×10 7 was plotted.Using the OD value reflected cell consistency and determined adhesion inhibition rate of Immune RNA.ResultsThe method hada good correlativity with counted cell LAI, r =0.96,and P <0.01,tha cofficients of variataion of reproducibiilty of intra , inter and different test assays were 6.3%~5.0% and 9.4% respectively.ConclusionThe method was superfine, simple and highly reproducible. It can be the method for determining the biological activity of immune RNA.
