天花粉热提取物抗柯萨奇B组病毒感染的研究

【摘要】 目的从体内、体外两方面对中药天花粉热提取物的毒性及抗病毒作用进行研究。 方法采取HeLa细胞系 ,在 96孔培养板上直接观察细胞病变 ;建立病毒性心肌炎小鼠模型 ,模型鼠经腹腔给予药物 ,记录小鼠的死亡情况 ,并从心肌中分离病毒 ,观察药物对病毒的清除作用 ;利用3 H -UR标记CVB3m,对其作用机理作初步探讨。结果天花粉热提取物在高浓度时具有一定细胞毒性 ,在较低浓度时具有明显的抑制病毒致细胞病变效应的作用 ;天花粉热提取物对Balb/c鼠具有一定毒副作用 ,但小鼠耐受良好 ,并对病毒感染小鼠具有一定保护作用 ;天花粉热提取物可作用于病毒的吸附和穿入过程。 结论天花粉热提取物具有抗病毒的临床应用潜力。

【Abstract】 Objective To study the effect of the extract of radix trichosanthes obtained by the heating and purifying technique on cytotoxicity and anti-CVB 3m infection in vitro and in vivo . Methods The HeLa cell strain was cultured in a flat-bottomed 96-well plastic microtiter tray to observe cytopathogenicity (CPE) directly. A infected CVB 3m animal model was set up. The infected mice were tested to evaluate the effect of the extract by administration of the extract into the abdominal cavity. The death time of the model mice was noted. The mechanism of the extract was elucidated using 3H-UR labeled CVB 3m . Results The extract had some cytotoxicity toward HeLa cells at a high concentration, and it obviously inhibited CPE at a lower concentration. The extract had toxic or side effect on Balb/c mice to a certain extent, although all mice could tolerate. The extract had obvious protective effect. The adsorption and penetration process of the virus was affected by the extract. Conclusion The extract has latent anti-virus capability.
