5种提取恶性疟原虫DNA方法用于PCR的结果评价

【摘要】 目的 比较 5种DNA模板制备方法的优缺点。 方法 采用 5种方法所制备的DNA进行PCR扩增 ,经统计学处理比较制备的成功率 ,与 6对不同引物PCR反应的敏感性 ,方便性 ,快速和费用等因素进行综合分析。结果 用于PCR ,方法 1~ 3的成功率显著高于法 4和 5 (P <0 .0 0 1) ;法 3制备的DNA敏感性最高 ,适用性最强。 结论  5种方法各有其优缺点 ,应根据实验目的的选用。

【Abstract】 Objective To compare the advantages and disadvantages of five common preparing DNA methods at present. Methods DNA template prepared by five methods was used for different Polymerase Chain Reaction(PCR).Successful preparing rates(SPRs),and sensitivity and practicability for PCR were compared by statistics analysis.Result SPRs of method 1,2 and 3 were significantly higher than those of methods 4 and 5.The PCR sensitivity was the highest when DNA prepared by mehtod 3 was used.Conclusion Every one of the five methods has its own advantages and disadvantages.They should be selectively used for different purposes.
