人乳腺癌HPV感染的超微结构和DNA分子原位杂交研究
Identification of human papillomaviruses in breast cancer by electron microscopy and DNA in situ hybridization
【摘要】 目的 :对乳腺癌的HPV感染进行形态学定位研究。方法 :对 4 6例乳腺癌切除标本及 7例良性病变标本进行透射电子显微镜以及HPVDNA分子原位杂交检测。结果 :透射电镜下 ,14例 (30 4 0 % )乳腺癌细胞核内有HPV样病毒颗粒 ,颗粒直径 30~ 5 0nm ,或弥漫散布 ,或聚集成团呈假结晶状排列。颗粒圆形或略不规则 ,含有HPV样颗粒的癌细胞核异型性较明显。7例良性病变细胞核内未见到HPV样颗粒。HPVDNA分子原位杂交显示 ,2 1例 (45 6 5 % )乳腺癌细胞核内HPV31/ 33DNA阳性 ,1例软骨肉瘤样化生性癌细胞核同时有HPV31/ 33及HPV16 / 18阳性 ;1例纤维腺瘤 (14 2 9% )也显示HPV31/ 33DNA阳性 (χ2 =2 74 31,P <0 0 5 )。电镜下 85 %病毒样颗粒阳性病例中可检测到HPVDNA存在 ,两种检测结果符合率达73 91%。结论 :乳腺肿瘤内存在未组装的、裸型病毒样颗粒。该研究对阐明乳腺癌的发病原因以及对乳腺癌的预防和治疗均有重要意义
【Abstract】 Purpose To observe the HPV in breast carcinoma morphologically. Methods Forty six cases of breast cancer and 7 cases of breast benign lesions had been studied by electron microscopy combined with HPV DNA in situ hybridization. Results HPV like virus particles were observed in 14 (30 40%) cases of breast carcinoma. The particles measured 30~50 nm in diameter, diffuse or aggregated as a pseudocrystal array. The cancer cells with HPV like particles disclosed obvious nuclear atypism. No HPV like particles were found in any case of benign lesions. Twenty one (45 65%) cases of breast carcinoma revealed HPV31/33 DNA positive by in situ hybridization method, one case of metaplastic carcinoma was both positive for HPV16/18 and HPV 31/33, and one case of fibroadenoma (14 29%) was also HPV31/33 DNA positive(χ 2 =2 7431, P <0 05). Eighty five percent of cases with HPV like particles existed HPV DNA in their nucei, the concordant rate of two methods reached 73 91%. Conclusions HPV can be morphologically localized in breast carcinoma by electron microscopy and in situ hybridization methods. It will be helpful for expounding the etiology of breast carcinoma.
