重组梅毒螺旋体表位抗原及多表位嵌合抗原研究

【摘要】 目的 :为梅毒检测试剂提供抗原性强、特异性高的重组表位抗原和多表位嵌合抗原。方法 :对梅毒螺旋体的主要抗原蛋白TpN15 ,TpN17,TpN44 .5和TpN47的优势抗原表位进行计算机分析 ,从中选取具有强抗原性的表位。根据大肠杆菌的偏性密码子 ,推断出这些抗原表位的基因序列 ,采用化学合成法合成表位抗原基因并克隆表达。结果 :获得了梅毒各抗原蛋白的优势抗原表位 ,构建了各表位基因及多表位嵌合基因的表达质粒 ,在E .coli中获得了高效表达。用获得的重组蛋白质纯品 ,对 12 2份心磷脂测定阳性血清样品进行测定 ,证明重组梅毒表位抗原和多表位嵌合抗原具有预期的抗原活性。结论 :重组梅毒表位抗原及多表位嵌合抗原具有较好的抗原活性 ,可用于研究不同类型的梅毒检测试剂

【Abstract】 Objective:To develop the strong and high specific epitope antigens and multi epitope antigen for use in serological diagnosis of syphilis.Methods:Four major Treponema pallidum proteins: TpN15, TpN17, TpN44.5 and TpN47 were analyzed by computer, and the immuno dominant epitopes were chosen. The genes of these chosen epitopes were synthesized according to the predominant codes in E.coli . The expression plasmids for these genes as well as an expression plasmid for a chimeric gene were constructed in vector pBVIL1 separately. The activity of expressed recombinant antigens was tested against a panel of sera with ELISA.Results:All the cloned genes of chosen epitopes and their chimeric genes were highly expressed in pBVIL1 in E.coli . The activity of all these recombinant antigens were high as expected.Conclusions:The activity of the recombinant epitope antigens of T.pallidum are high and specific,so the recombinant epitope antigens are suitable for use in serological diagnosis of syphilis.
