肝癌相关基因差异表达分析
Analyses of differential expression of hepatocellular carcinoma-related genes
【摘要】 为了研究肝癌发生和发展的分子机理 ,用cDNAMicroarray技术比较肝癌 (HCC)和癌旁组织 (nonHCC)基因差异表达情况 ,并选择 4个差异表达基因用RT PCR验证。在 1176个肿瘤相关基因中 ,4 91个在HCC中表达 ,345个在nonHCC中表达。HCC中表达上调的基因 172个 ,表达下调的基因 89个 ,RT PCR结果与cDNAMicroarray的结果基本一致。差异表达最多的基因是与细胞信号传导和细胞分裂相关的基因。肝癌中生长因子及其受体相关基因表达上调 ,Ras Raf MAPK信号传导途径活化 ,与细胞周期相关基因的异常表达加速了细胞分裂进程。所有这些变化与肝癌的发生和发展密切相关
【Abstract】 To understand the molecular mechanism of hepatocarcinogenesis we compared gene expression changes between hepatocellular carcinoma(HCC) and non cancer liver tissue(nonHCC) by cDNA microarray. Of the 1176 cancer related genes, 491 genes were expressed in HCC and 345 genes in nonHCC. 172 genes were up regulated while 89 genes were down regulated in HCC. Many differentially expressed genes were involved in cell signaling and cell division. Up regulated growth factors and their receptors , the actived Ras Raf MAPK pathway and deregulated cell cycle related genes may contribute to hepatocarcinogenesis.
