移植的心肌细胞在心梗后疤痕组织中存活的研究
Survival possibility of grafted cardiomyocytes colonize myocardial infarct area
【摘要】 目的 探讨心肌细胞移植到正常和心梗后疤痕心肌中生存的可行性。方法 通过结扎冠状动脉前降支制成心梗模型 ,利用 (18± 2 )天的孕鼠心室肌细胞经培养、标记后移植到大鼠正常和心梗后疤痕心肌组织中 ,4周后取病理做HE和免疫组化染色。结果 HE和免疫组化染色显示 ,移植细胞在大鼠正常和疤痕心肌细胞中分化、增殖良好 ,可见横纹和核内结构 ,形成了富含毛细血管与宿主心肌有序排列的新生组织结构。结论 说明了移植同种心肌细胞到心梗后疤痕心肌组织中生存增殖的可行性和改善心功和血供的先决条件。
【Abstract】 Objective To observe whether allogenic cells can graft into myocardial infarct(MI) area and survive there.Methods Neonatal cardiomyocytes were obtained from fetuses of female rats (18±2)days pregnant,then injected into the left ventricular(LV) myocardium of normal rats or into the left ventricular MI area of rats in which MI had been created by proximal occlusion of the left coronary artery,control rats were injected with the culture medium alone.After four weeks,LV slices were processed for histological (hematoxylin eosin) and immunocytochemical labeling techniques with monoclonal anti bromocleoxyuridine(BrdU).Results Examination of serial sections from injuected rejoins showed that grafted myocytes were harbored and lived in the host LV myocardium in 8 normal animals and M1 area in 7 the infarcted rats.Grafted cells were identified by their morphological and immunocytochemical characteristics.They had striated and nucleocytopiasmic structures and lined with host myocardial tissue orderly,which formed a newborn myocardium with many blood capillaries and less lymphocytes.Conclusion These initial results suggest the feasibility of transplanting allogeneic cardiomyocytes into the MI areas and a prerequisite for this approach to successfully improve the function and supply blood flowing into ischemically damaged myocardium.
