三氧化二砷诱导骨肉瘤MG-63细胞凋亡的实验研究
Experimental study on the apoptotic effect of arsenic trioxide on human osteosarcoma MG-63 cells
【摘要】 目的 :探讨三氧化二砷 (As2 O3)诱导骨肉瘤MG 6 3细胞凋亡的作用。方法 :用MTT法、透射电镜、琼脂糖凝胶电泳等方法观察As2 O3诱导MG 6 3细胞凋亡的作用。结果 :As2 O3可有效地抑制MG 6 3细胞增殖 ,随着药物浓度增高其抑制作用增强 ,且细胞出现典型的凋亡变化。DNA琼脂糖凝胶电泳出现凋亡特征性梯形条形。透射电镜下可见细胞核仁消失 ,染色质浓缩聚集于核膜下。结论 :As2 O3可诱导骨肉瘤细胞凋亡 ,其作用机制有待进一步探讨
【Abstract】 Objective To study the apoptotic effect of arsenic trioxide(As 2O 3)on human ostrosarcoma MG 63 cells. Methods The MTT reduction test, transmission electron microscopy and agarose gel electrophoresis were applied to observe the apoptotic changes of human osteosarcoma line MG 63 induced by As 2O 3. Results Typical apoptotic cellular changes were found after As 2O 3 treatment. Transmission electron miroscopy showed that nucleolus disappeared and chromatin condensed under the nucleonic membrane. Agarose gel electrophorsis showed a "ladder" strand of DNA, a special phenomenon of cellular apoptosis. The above mentioned changes were not found in the control group.Conclusion As 2O 3 can induce apoptosis of human osteosarcoma MG 63 cells. Further studies are to be carried out on its mechanism.
