侧脑室注射红霉素对糖尿病大鼠胃窦运动的影响
EFFECTS ON GASTRIC MOTILITY IN DIABETIC RATS BY INJECTING ERYTHROMYCIN INTO LATERAL VENTRICLE OF CEREBRUM
【摘要】 ①目的 探讨红霉素 (EM)对糖尿病模型大鼠胃窦运动的中枢作用机制。②方法 应用清醒大鼠胃窦运动记录技术 ,观察了侧脑室微量注射EM对糖尿病模型大鼠胃窦运动的影响。将实验大鼠随机分为 4组 :对照组 (A组 )侧脑室注射生理盐水 2 μL ;B组侧脑室注射EM 5 0 μg ;C组侧脑室注射EM 10 0 μg ;D组侧脑室注射GM 10 9+EM .③结果 B组 5min时胃收缩幅度变化率与A组比较差异有显著性 (F =16 .4 2 ,q =2 .4 6 ,P <0 .0 5 ) ;C组 5min时胃收缩幅度变化率与A组比较差异有显著性 (F =16 .4 2 ,q =3.5 2 ,P <0 .0 5 ) ,10min时差异有显著性 (F =15 .71,q =3.38,P <0 .0 5 ) ,15min时差异有显著性 (F =8.5 5 ,q =3.12 ,P <0 .0 5 ) ;C组 5min时胃收缩幅度变化率与B组比较差异有显著性 (F =11.2 4 ,q =3.2 5 ,P <0 .0 5 )。B组 5min时胃收缩频率增加与A组比较差异有显著性 (F =11.2 4 ,q =3.12 ,P <0 .0 5 ) ;C组 5min时胃收缩频率与A组比较 ,差异有显著性 (F =11.2 4 ,q =3.0 2 ,P <0 .0 5 ) ,10min时差异有显著性 (F =14 .18,q =3.6 0 ,P <0 .0 5 ) ,15min时差异有显著性 (F =13.77,q =3.4 9,P <0 .0 5 )。侧脑室给予胃动素受体拮抗剂GM 10 9(2 μg)可阻断EM(10 0 μg)对胃窦运动幅度和收缩频率的影响。 ④结论
【Abstract】 Objective\ To study the mechanism mediating the effects of erythromycin(EM) on gastric motility in diabetic model rats.\ Methods\ The influence of intra cerebroventricle micro injection (i.c.v.) of EM on gastric motility in conscious rats was recorded. The rats were randomly divided into 4 groups: Group A(control group), normal saline 2 μL, injected into the lateral ventricle of cerebrum; Group B, EM 50 μg; Group C, EM 100 μg and Group D, GM 109+EM.\ Results\ The amplitude of gastric motility of Group B was higher than that of Group A 5 minutes after the injection, the difference being significant ( F= 16.42, q=2.46, P <0.05). The differences of amplitude between Group C and Group A were significant at 5 minutes ( F= 16.42 , q= 3.12 , P <0.05), 10 minutes ( F=15.71, q=3.38, P <0.05) and 15 minutes ( F=8.55, q=3.12, P <0.05) after the injection .The amplitude of group C was higher than that of group B 5 minutes after the injection ( F=11.24, q=3.25, P <0.05). The frequency of gastric motility of Group B was higher than that of group A 5 minutes after the injection ( F=11.24, q=3.12, P < 0.05 ). The frequency of Group C was higher than that of Group A 5 minutes ( F=11.24, q=3.02, P <0.05), 10 minutes ( F= 14.18 , q=3.60, P <0.05), and 15 minutes( F=13.77, q=3.49, P <0.05) after the injection, the differences being significant. The i.c.v. of GM 109 2 μg could block the effects of 100 μg of EM on the amplitude and frequency of gastric motility.\ Conclusion The central administration of EM to enhance gastric motility may be related to the mediation of the central motilin receptor.
