“肾应冬”调控机制的分子生物学实验研究
Experimental Research of Molecular Biology on Modulation Mechanism of Correspondence between Kidney and Winter
【摘要】 目的 :“肾应冬”是中医基础“五脏应时”理论的一个部分。方法 :采用大鼠松果体摘除模型 ,运用原位杂交技术 ,探讨了在冬至、夏至SD雄性大鼠睾丸c fos和c jun的mRNA的变化规律。结果 :睾丸c fos和c jun的mRNA具有冬至表达强、表达量多 ,夏至表达弱、表达量少的特点和趋势。摘除松果体在冬至使睾丸c fos和c jun的mRNA明显减弱 ,而在夏至则使睾丸c fos和c jun的mRNA明显增强。结论 :1 “肾应冬”在生殖方面的调控机制可能是 ,通过对肾所藏的生殖之精的两种调控成分即促进生殖之精的物质和抑制生殖之精的物质而起作用。肾封藏的抑制生殖之精的物质和功能在冬季加强 ,而所储藏的促进生殖之精的物质和功能在冬至减弱 ,故在冬至 ,肾外泻生殖之精的功能减弱 ,生殖机能处于低潮。 2 “肾应冬”的调控机制是肾中精气随季节的变化以松果体为中介 ,通过影响睾丸的c fos和c jun的mRNA表达来调节季节性生殖活动。 3 中医“肾应冬”具有客观物质基础 ,中医“以时测脏”具有正确性和科学性。
【Abstract】 Objective:Correspondence between Kidney and Winter is one part of Correspondence between five viscus and the five seasons of Basic Theory of TCM. Adopting rat pinealectomy model and in situ hybridization technology, we discuss the varying regularity of c fos and c jun mRNA in male rat’s testis in midwinter and in midsummer. Results:the expression degree and the amount of c fos and c jun mRNA in testis are of intensity and plenty feature and trend in midwinter while weak and insignificant in midsummer. After pinealectomy, the expression degree and the amount of them decend significantly in midwinter while in midsummer rise obviously. Conclusion:1.The modulation mechanism expressed in reproduction of Correspondence between Kidney and Winter may function through two kinds of modulation elements the materials of promoting and restricting reproductive essence stored in the kidney. The latter strenghen their action meanwhile the former weaken their action in midwinter, so the kidney weaken its function of excrete reproductive essence, as a result the reproductive power of kidney descend.2.The modulation mechanism of the Correspondence between Kidney and Winter is that the essence and Qi of kidney varies according to the season’s change which is mediated by pineal to influence c fos and c jun mRNA in testis to modulate season’s reproductive activity. 3. Correspondence between Kidney and Winter of TCM is based on objective material. Theory of researching viscera through seasons’ changes is correct and scientific.
