建立Duchenne型肌营养不良鼠的骨髓移植模型

【摘要】 【目的】建立Duchenne型肌营养不良鼠 (mdx鼠 )的骨髓移植模型 ,摸索有效的放疗剂量。【方法】用体外培养的不同数量的C5 7BL/ 6雄鼠的骨髓细胞、骨髓基质细胞、骨髓悬浮细胞 ,用不同方法移植给不同剂量放疗后的mdx鼠 ,观察受体鼠的存活率及存活时间 ,并对死亡鼠作GVHD(移植物抗宿主病 )的病理学检测。【结果】 16只mdx鼠移植前 3d 12Gyγ射线照射 ,在移植后 7d内全部死亡 ,且注射细胞数较多者 ,存活时间相对较长 ;8只经 6Gyγ射线照射的mdx鼠 ,移植后 6 0d ,存活 4只 ,其中 1例经腹腔注射骨髓细胞、1例肌肉局部注射骨髓细胞 ,移植后 7d内死亡 ;而 8Gyγ射线照射的 14只mdx鼠 ,移植后 6 0d 10例存活 ,4例死亡。【结论】本实验条件下 ,移植前 8Gyγ射线放疗较合理 ,在有效剂量放疗破坏受体骨髓造血系统后 ,静脉注射移植同种鼠的骨髓细胞、基质细胞、悬浮细胞 ,mdx鼠存活率较高 ,且移植细胞数量以 1× 10 7个 /只鼠为佳。

【Abstract】 Objective To establish an marrow transplantation model in mice with Duchenne muscular dystrophy(mdx), in order to study the effective irradiation dose. Methods Different amounts of bone marrow cells, stromal cells, suspension cells were cultured in vitro from C57BL/6 male mice, and the cells were transplanted into the irradiated mdx mice with different means. The survival rate and survival time were investigated. Histopathology of GVHD (graft versus host disease) was assessed for the died mice. Results 16 mdx mice irradiated with 12 Gy γ ray 3 d before transplantation died within 7 d, and the mice injected with more cells lived longer; 8 mice irradiated with 6 Gy, 4 of them survived 60 d after transplantation, one of them was transplanted by intraperitoneal injection with bone marrow cells, another was injected by local muscle, both of them died within 7 d after transplantation; 14 mice irradiated with 8 Gy, 10 of them survived 60 d after transplantation, 4 died. Conclusion This study shows that 8 Gy γ ray is the best effective irradiation dose. After the hematopoietic system is destroyed by effective irradiation, mdx mice are able to survive longer after injection with syngeneic bone marrow cells, stromal cells, and suspension cells, and the better survive rate can be achieved by injection of 1×10 7 transplantation cells for each mouse.
