受体外周血IgG、MΦ、NK、CD4、CD8及MLR的变化与异种移植物存活的关系
Relation between the xenograft survival and IgG, MΦ, NK,CD4,CD8, MLR testing in peripheral blood of the recipient
【摘要】 目的 :探讨受体外周血IgG、MΦ、NK、CD4、CD8及MLR的变化与异种移植物存活的关系。方法 :选用小鼠移植于大鼠耳后心肌组织模型 ,按实验所设Ⅰ、Ⅱ、Ⅲ、Ⅳ组 :在术前 12、8、4、0d及 10、6、2、0d分别将小鼠脾细胞 1× 10 8个及抗脾细胞血清 0 2ml静注大鼠 ,术日始CsA 10mg (kg·d) ,Cy 2 0mg (kg·d) ,CD4McAb、CD8McAb、MΦMcAb、NKmAb均为 2 5 0 μg (kg·d) ,CCV 0 2mg (kg·d)腹腔注射至术后 6d。结果 :Ⅱ、Ⅲ、Ⅳ组移植小鼠心肌存活延长 ,尤以Ⅳ组存活时间达 2 1d左右 (P <0 0 5 ) ,术后 7d外周血MΦ、NK细胞显著增多 (P <0 0 1) ,并维持至术后 2 1d ;移植术后 7dCD4、CD8稍增多 ,术后 2 1d显著增加 (P <0 0 5 ) ;术后 7dⅡ、Ⅲ、Ⅳ组IgG显著下降 (P <0 0 5 ) ;术后 7,2 1d动态MLR比较无显著差异 (P >0 0 5 )。结论 :外周血IgG、MΦ、NK细胞的变化可作为监测异种移植细胞性排斥反应的指标 ,异种移植物存活时间延长 ,将面对宿主T细胞免疫应答。
【Abstract】 Objective:To study the relation between the xenograft survival and IgG, MΦ, NK,CD4, CD8,MLR testing in peripheral blood of the recipient Methods:By using mouse to rat heart tissue transplantation model,4 experimental groups were established and treated by using: CCV, RPMI 1640(Ⅰ);splenocyte, anti serum with CCV therapy (Ⅱ); splenocyte, anti serum with CsA, Cy, CCV therapies (Ⅲ);splenocyte, anti serum with CsA, Cy, CCV, anti MΦ, anti NK, anti CD4 and anti CD8 therapies (Ⅳ) The rats received splenocytes (1×10 8) intravenously on the day -12, -8, -4, 0;anti serum (0 2 ml) on the day -10, -6, -2, 0, and CsA 〔10 mg/(kg·d) i p〕, Cy 〔20 mg/(kg·d) i p〕, CCV 〔0 2 mg/(kg·d) i p〕, anti CD4,anti CD8,anti MΦ and anti NK 〔250 μg/(kg·d)〕 on the day from 0 to 6 relative to heart grafting on day 0 The graft survival, the IgG, CD4,CD8, MΦ, NK, MLR were studied Results: On the 7th day after transplantation, grafts in groupⅠwere destroyed, but the grafts in group Ⅱ, Ⅲ, Ⅳ survived well; on the 21st day, only grafts in group Ⅳsurvived well (P<0 05 vs group Ⅱ, Ⅲ) The trend analysis of MLR on the 7th and 21st posttransplantative days showed no positive correlation The serum IgG values in group Ⅱ,Ⅲ,Ⅳ decreased significantly on the 7th day (P<0 05,vs groupⅠ,P<0 05 vs those on the day 0 and on the 21st day) The result of CD4,CD8 in peripheral blood increased from on the 7th day to 21st day (P<0 05)and the result of MΦ, NK in peripheral blood reached a peak on the 7th day Conclusion: The results of MΦ, NK, IgG testing in peripheral blood can act as predictors for xenograft rejection and the results of CD4,CD8 testing in peripheral blood implicated that T cells may play an important mediating roles in the rejection.
