化疗药物诱导急性白血病原代细胞凋亡的研究

【摘要】 目的 :探讨化疗药物诱导急性白血病 (AL )原代细胞凋亡的时间、剂量、效应关系。方法 :以成人 AL原代细胞为研究对象 ,采用高、中、低 3种浓度梯度 ,模拟人体应用 HHar、Ara- C和 ACR后的最大血液浓度 ,应用一系列经典方法检测细胞凋亡 ,并分析得出相应的时间、剂量、效应关系。结果 :HHar、Ara- C具有时间 -效应及剂量-效应线性趋势 ,而 ACR低剂量组诱导细胞凋亡的能力大于高剂量组 ,并具有时间 -效应线趋势 ;高剂量组 HHar、Ara- C诱导细胞凋亡的能力大于高剂量组 ACR,而低剂量组 ACR诱导细胞凋亡的能力大于低剂量组 HHar及Ara- C;3种药物对 AML原代细胞诱导凋亡的效果均优于对 AL L原代细胞。结论 :AL原代细胞对细胞凋亡的敏感性也是决定化疗效果的关键因素

【Abstract】 Objective: To explore the time dose effect relationships of chemotherapeutic agents in inducing primary acute leukemia cells apoptosis .Methods:Primary acute leukemia cells from 34 patients were treated in virto with three widely used chemotherapeutic agents:homoharringtonium(HHar),cytarabinum(Ara C) and aclarubicinum (ACR) of different concentration gradients. A series of classical methods were used to detect apoptoses.The time dose effect relationships were analyzed furthermore.Rusults:The apoptotic rates and DNA fragmentation ratios of the primary leukemia cells treated with HHar or Ara c had the time effect and dose effect linear trend.However,the low dose group of ACR had a stronger ability of inducing apoptosis than the high dose one.The apoptotic rates and DNA fragmentation ratios of the low dose group of ACR had the time effect linear tendency.Comparing the inducing apoptosis ability of the three agents,it Was found that the high dose group of HHar and Ara C and low dose group of ACR could produce more primary leukemia cells apoptosis.The AML cells were more sensitive to the apoptosis induced by HHar or ACR or Ara C than the ALL ones. Conclusion:The apoptosis inducing effect of a certain chemotherapeutic agent to the primary AL cells may be related to its therapeutic effect in the treatment of AL.
